Delivering functional nucleic acids to mammalian cells via bacterially-derived, intact minicells

Abstract

Intact bacterially derived minicells containing functional nucleic acids or plasmids encoding functional nucleic acids can reduce, in targeted mammalian cells, drug resistance, apoptosis resistance, and neoplasticity, respectively. Methodology that employs minicells to deliver functional nucleic acids, targeting the transcripts of proteins that contribute to drug resistance or apoptosis resistance, inter alia, can be combined with chemotherapy to increase the effectiveness of the chemotherapy.

    A composition comprising (a) an intact, bacterially derived minicell that contains a functional nucleic acid molecule or a plasmid comprising a segment that encodes a functional nucleic acid molecule and (b) a pharmaceutically acceptable carrier therefor, wherein said functional nucleic acid molecule targets the gene or the transcript of a protein that contributes to resistance to a chemotherapeutic drug.

    A chemotherapeutic drug for use in the treatment of a drug resistant malignancy, wherein an intact, bacterially derived minicell containing a functional nucleic acid molecule or a plasmid comprising a segment that encodes a functional nucleic acid molecule, where the functional nucleic acid molecule targets the gene or transcript of a protein that promotes resistance to said chemotherapeutic drug, is administered simultaneously or consecutively with said chemotherapeutic drug to allow the functional nucleic acid to diminish resistance to said chemotherapeutic drug.

    A product containing a chemotherapeutic drug and an intact, bacterially derived minicell, said minicell containing a functional nucleic acid molecule or a plasmid comprising a segment that encodes a functional nucleic acid molecule, where the functional nucleic acid molecule targets the gene or transcript of a protein that promotes resistance against said drug, for use in the treatment of a malignancy resistant to said chemotherapeutic drug, wherein said drug and intact minicell is administrated simultaneously or consecutively.

    The intact, bacterially derived minicell, the chemotherapeutic drug or the product according to any of the preceding claims, wherein said malignancy resistant to said chemotherapeutic drug is selected from the list consisting of cancer, AIDS and tuberculosis.

    The intact, bacterially derived minicell, the chemotherapeutic drug or the product according to any of the preceding claims, wherein said minicell further comprises a bispecific ligand.

    The intact, bacterially derived minicell, the chemotherapeutic drug or the product according to any of the preceding claims, wherein said bispecific ligand is selected from the list consisting of receptors, enzymes, binding peptides, fusion/chimeric proteins and small molecules.

    The intact, bacterially derived minicell, the chemotherapeutic drug or the product according to any of the preceding claims, wherein said bispecific ligand has specificity for both a surface component on the minicell and a surface component on a non-phagocytic mammalian cell.

    The intact bacterially derived minicell, the chemotherapeutic drug or the product according to any of the preceding claims, wherein the target of said minicell is a phagocysis- or endocytosis-competent mammalian cell.

    The intact, bacterially derived minicell, the chemotherapeutic drug or the product according to any of the preceding claims for administration at least once a week over several weeks to several months.
