Abstract

A system for processing the contents of one or more receptacles to detect an optical signal, if any, emitted by the contents of each receptacle, the system comprising: an incubator including a temperature-controlled chamber configured to receive one or more receptacles; at least one signal detector configured to detect an optical signal, if any, emitted by the contents of a receptacle disposed within said chamber in an operative position with respect to said signal detector; and at least one magnet disposed within said chamber so as to be adjacent to a receptacle that is in the operative position with respect to said signal detector.

    A system for processing the contents of one or more receptacles to detect an optical signal, if any, emitted by the contents of each receptacle, the system comprising:an incubator including a temperature-controlled chamber configured to receive one or more receptacles;at least one signal detector configured to detect an optical signal, if any, emitted by the contents of a receptacle disposed within said chamber in an operative position with respect to said signal detector;and at least one magnet disposed within said chamber so as to be adjacent to a receptacle that is in the operative position with respect to said signal detector.

    The system of any one of claims I to 5, wherein each signal detector comprises a fluorometer configured to detect a fluorescent signal, if any, emitted by the contents of a receptacle operatively positioned with respect to said fluorometer.

    The system of any one of claims I to 7, wherein the system is configured to process detected optical signal data so as to determine the presence or amount of a target nucleic acid within the contents of each receptacle based on the detected optical signal.

    The system of any one of claims 1 to 8, further comprising a separation station having movable magnets for selectively exposing the contents of a receptacle within said separation station to a magnetic field, thereby isolating magnetically-responsive solid supports within the receptacle.

    An automated method for processing the contents of multiple reaction receptacles to determine the presence or amount of a target nucleic acid in each of the reaction receptacles, the method comprising the steps of:a) isolating and purifying target nucleic acid that may be present in a sample material contained within each of the reaction receptacles by: i) exposing each reaction receptacle to a magnetic field, thereby isolating magnetically-responsive solid supports within each reaction receptacle, the solid supports being capable of directly or indirectly immobilizing the target nucleic acid thereon;ii) during step i), removing at least a portion of the sample material in each reaction receptacle from the isolated solid supports;and iii) after step ii), removing the magnetic field and washing the solid supports in each reaction receptacle with a wash solution;b) moving the reaction receptacles to a temperature-controlled chamber of an incubator, each reaction receptacle being provided with reagents sufficient to amplify and detect the presence of a target sequence contained within the target nucleic acid under the temperature conditions of the incubator;c) exposing each reaction receptacle to a magnetic field within the incubator, thereby aggregating at least a portion of the solid supports within each reaction receptacle;and d) during step c), measuring the amount of a signal associated with an amplification product containing the target sequence or its complement in each reaction receptacle provided to the incubator in accordance with step b).

    The method of any one of claims 11 to 13, wherein the incubator includes a reaction receptacle carrier that is rotatable about an axis of rotation and is configured to receive and hold reaction receptacles moved to the incubator.

    The method of any one of claims 11 to 17, wherein step (c) comprises placing the reaction receptacle in proximity to at least one magnet configured to aggregate at least a portion of the solid supports within the reaction receptacle.

    The method of any one of claims 11 to 19, wherein step d) is performed periodically on each reaction receptacle, and wherein the method further comprises:e) quantifying the amount of the target nucleic acid present in each reaction receptacle based upon the measurements obtained in step d);and f) successively removing reaction receptacles having contents processed in accordance with steps c)-e) from the incubator, wherein reaction receptacles are being successively moved to the incubator in accordance with step b) as the contents of reaction receptacles previously provided to the incubator are being processed in accordance with steps c)-e).
