Abstract

A method for reducing a nucleic acid amplification inhibitor in a receptacle comprises the steps of coating a portion of an inner surface of a receptacle with a surface treating agent, such as an oil, that reduces the amount of an amplification inhibitor adhering to the inner surface during a process of isolating a target nucleic acid within the receptacle, washing the target nucleic acid, and amplifying a target sequence of the target nucleic acid. The surface treating agent does not inhibit amplification of the target sequence.

A method for reducing the presence of a nucleic acid amplification inhibitor in a receptacle, said method comprising the steps of: (a) providing a nucleic acid containing material to a receptacle having an inner surface;(b) after step (a), coating that portion of the inner surface of the receptacle that is in contact with the nucleic acid containing material with a surface treating agent, the surface treating agent being an oil that reduces the amount of an amplification inhibitor that can adhere to the inner surface of the receptacle;(c) after step (b), exposing a first volume of the receptacle comprising the nucleic acid containing material to reagents and conditions sufficient to isolate a target nucleic acid;(d) removing at least a portion of the first volume from the receptacle;(e) after step (d), providing to the receptacle a wash solution;(f) exposing a second volume of the receptacle comprising the wash solution to reagents and conditions sufficient to isolate the target nucleic acid;(g) removing at least a portion of the second volume from the receptacle;and (h) after step (g), providing to the receptacle reagents sufficient to amplify a target sequence of the target nucleic acid, and exposing the receptacle to amplification conditions, wherein the surface treating agent will not inhibit amplification of the target sequence.

    The method of any one of claims 1 to 3, wherein the surface treating agent is a silicone oil.

    The method of any one of claims 1 to 4, wherein step (b) comprises agitating the receptacle.

    The method of any one of claims 1 to 5, wherein the reagents sufficient to isolate the target nucleic acid comprise a solid support material for immobilizing the target nucleic acid.

    The method of any one of claims 1 to 7, wherein the first and second volumes are exposed to a magnetic field in steps (c) and (f).

    The method of any one of claims 1 to 10, wherein the receptacle comprises a hydrophobic material.

    The method of any one of claims 1 to 11, wherein the receptacle comprises polypropylene.

    The method of any one of claims 1 to 12, wherein the amplification inhibitor is provided by the nucleic acid containing material.

    The method of any one of claims 1 to 16, wherein the reagents for amplifying the target sequence include a primer, nucleoside triphosphates, and a nucleic acid polymerase capable of extending the 3' end of the primer hybridized to the target nucleic acid.

    A method for reducing the presence of a nucleic acid amplification inhibitor in a receptacle, the method comprising:(a) removing at least a portion of a first volume of a receptacle having an inner surface, the first volume comprising a nucleic acid containing material;(b) after step (a), providing to the receptacle a wash solution and a surface treating agent, wherein the surface treating agent is not an inhibitor of a nucleic acid amplification reaction;(c) coating that portion of the inner surface of the receptacle that is in contact with the wash solution with the surface treating agent, the surface treating agent being an oil that reduces the amount of an amplification inhibitor that can adhere to the inner surface of the receptacle;(d) after step (c), exposing a second volume of the receptacle comprising the wash solution to reagents and conditions sufficient to isolate a target nucleic acid;(e) removing at least a portion of the second volume from the receptacle;and (f) after step (e), providing to the receptacle reagents sufficient to amplify a target sequence of the target nucleic acid, and exposing the receptacle to amplification conditions, wherein the surface treating agent will not inhibit amplification of the target sequence. - 131 CA 02842232 2014-02-06
