15 claims: 4 independent, 11 dependent

  1. 1
A method of modifying a trait of a selected plant comprising:(a) stably transforming cells from a selected plant using a recombinant DNA molecule that comprises (i) a T-DNA border-like sequence and (ii) a desired polynucleotide, wherein (I) the desired polynucleotide consists of nucleic acid sequences that are native to the selected plant, native to a plant from the same species, or native to a plant that is sexually interfertile with the selected plant, and (II) wherein the T-DNA border-like sequence is not identical in nucleotide sequence to any known Agrobacterium-derived T-DNA border sequence, but wherein the T-DNA border-like sequence promotes and facilitates the integration of the desired polynucleotide;and (b) obtaining a stably transformed plant from the transformed plant cells containing the desired polynucleotide stably integrated into the genome, wherein expression of the desired polynucleotide modifies a trait of the plant.
  2. 5
The method of claims 4, wherein (a) the monocotyledenous plant is selected from turf, turf grass, cereal, maize, rice, oat, wheat, barley, sorghum, orchid, iris, lily, onion, banana, sugarcane, and palm;and wherein (b) the dicotyledenous plant is selected from potato, tobacco, tomato, sugarbeet, broccoli, cassava, sweet potato, pepper, cotton, poinsettia, legumes, alfalfa, soybean, carrot, strawberry, lettuce, oak, maple, walnut, rose, mint, squash, daisy, and cactus.
  3. 12
A plant exhibiting a modified expression of a trait, said plant having stably integrated into its genome a recombinant DNA molecule consisting of genetic elements that are (i) native to the selected plant, (ii) native to a plant from the same species as the selected plant, or (iii) native to a plant that is sexually interfertile with the selected plant, wherein the recombinant DNA molecule modifies the expression of the trait and wherein said recombinant DNA molecule is introduced into plant cells using a P-DNA that has been modified to comprise, in the 5' and 3' direction:a) a first T-DNA border-like sequence that promotes and facilitates the integration of the desired polynucleotide, wherein the T-DNA border-like sequence is not identical in nucleotide sequence to any known Agrobacterium- derived T-DNA border sequence;b) a promoter that is not native to the desired polynucleotide but is (1) native to the selected plant, (2) native to a plant from the same species as the selected plant, or (3) native to a plant that is sexually interfertile with the selected plant;c) a desired polynucleotide that is operably linked to the promoter;d) a terminator;and e) a second T-DNA border-like sequence that promotes and facilitates the integration of the desired polynucleotide, wherein the T-DNA border-like sequence is not identical in nucleotide sequence to any known Agrobacterium- derived T-DNA border sequence.
  4. 15
The plant of any one of claims 12 to 14 that is a potato plant.
