    A system for collecting a desired blood component, the system comprising:a permanent tower system (2000) having a centrifuge chamber (2101);a photoactivation chamber and a fluid flow control deck (1200);a disposable photopheresis kit comprising: a cassette (1100) for valving, pumping and controlling the movement of blood fluids during a photopheresis treatment session;a centrifuge bowl (10) having an inlet port and two outlet ports;a first hematocrit sensor positioned in the centrifuge chamber;a treatment bag (50) coupled to the cassette by outlet tube (1116);a plasma collection bag (51);an irradiation chamber (700);wherein the photopheresis kit has the necessary tubing and connectors to fluidly connect all devices and route the circulation of fluids during a photopheresis treatment session;wherein the control deck includes a controller operably connected to the necessary components of the system (1210) and five peristaltic pumps (1301-1305), the controller programmed to: combine anticoagulant with whole blood withdrawn from a patient at a selected ratio of anticoagulant to whole blood;pump a combination of whole blood and anticoagulant through the cassette to the centrifuge bowl;operate the centrifuge bowl until air is displaced and to separate the mixture of whole blood and anticoagulant into a plurality of blood components according to density;and continue to pump the combined whole blood and anticoagulant into the centrifuge and collect separated plasma until a selected amount of plasma is collected;mix plasma with a priming fluid and return the mixture of plasma and priming fluid to the patient at the same rate as incoming whole blood until red blood cells are detected by the first hematocrit sensor positioned in the centrifuge chamber;withdraw red cells and pump the red cells at a controlled speed so as to maintain a red cell line selected level in the centrifuge bowl while buffy coat is collected in the centrifuge bowl;and, at a selected time, when a desired amount of buffy coat cells are collected in the centrifuge bowl, cause the pump to continue to pump whole blood into the centrifuge bowl while discontinuing pumping red blood cells, thereby causing the red blood cells to push buffy coat out of the centrifuge bowl through the outlet tube (1116) until a desired amount is collected in the treatment bag (50);and discontinue collection of buffy coat when red blood cells have been detected by a second hematocrit sensor (1125) operably connected to the outlet tube, mix a photoactivation chemical with the buffy coat to form a mixture of the photoactivation chemical and the buffy coat;pump the buffy coat and photoactivation chemical mixture from the treatment bag to the irradiation chamber;and irradiate the combination of the buffy coat and photoactivation chemical to produce an irradiated combination for return to the patient, wherein the entire photopheresis treatment is completed in less than 70 minutes.

    The system of any one of claims 2 to 4, wherein the controller is further programmed to separate the buffy coat cells and to collect one or more selected components, the components selected from the group consisting of platelets and leukocytes.

    The system of any one of claims 2 to 5, wherein the controller is further programmed to prime the centrifuge bowl with anticoagulant and saline mixture.

    The system of any one of claims 2 to 6, wherein the controller is further programmed to rinse the centrifuge bowl, treatment bag, and irradiation chamber with saline to provide a resultant rinse solution for return to the patient.
