Composition containing radioisotopes immobilised on solid particles, and its use in brachytherapy

Abstract

Radioactive composition for use in the therapy of neoplastic and non-neoplastic diseases by means of the application of radioactive sources in direct contact with the tumor tissues or within the same, and for use as an aid in radio-guided surgery, containing radioisotopes immobilized on biocompatible or bioabsorbable solid particles. The said solid particles are incorporated in a biocompatible and bioabsorbable matrix consisting of a hypotonic gel containing, in water, from 2 to 30% by weight of polyvinylpyvrrolidone and from 0.01 to 2% by weight of agar-agar or agarose. The invention also concerns a radiopharmaceutical kit or a kit for use in radio-guided surgery comprising, as separate components, the single constituents of the above composition.

    A radioactive composition for use in therapy and as a surgical aid, containing radioisotopes immobilized on biocompatible or bioabsorbable solid particles, characterised in that the said solid particles are incorporated in a biocompatible and bioabsorbable matrix consisting of a hypotonic gel containing, in water, from 2 to 30% by weight of polyvinylpyrrolidone and from 0.01 to 2% by weight of agar-agar or agarose.

    A composition according to any one of claims 1-3, wherein the said radioisotope-carrying biocompatible or bioabsorbable solid particles are obtainable from separately produced solid particles, on which the said radioisotopes are subsequently immobilized by chelation.

    A composition according to any one of claims 1-3, wherein the said radioisotope-carrying biocompatible or bioabsorbable solid particles are obtainable from particles that already incorporate the non-radioactive chemical element corresponding to the desired radioisotope, which particles are then irradiated in a nuclear reactor under neutron flux to obtain the said radioisotopes.

    A composition according to claims 4 or 5, wherein the said solid particles are chosen among bioinert particles made of ion exchange resins and bioabsorbable particles made of poly(lactic acid).

    A composition according to any one of claims 4-6, wherein the said particles have diameters in the range from 0.01 µm to 100 µm.

    A composition according to any one of the previous claims, wherein the said radioisotopes are beta- and/or gamma-emitting radioisotopes.

    A radiopharmaceutical kit or a kit for use in radio-guided surgery comprising a first component containing biocompatible or bioabsorbable solid particles capable of carrying radioisotopes in a substantially not leachable manner, and a second component consisting of a hypotonic gel containing, in water, from 2 to 30% by weight of polyvinylpyrrolidone and from 0.01 to 2% by weight of agar-agar or agarose.

    A kit according to any one of claims 11-13, wherein the average molecular weight (MW) of said polyvinylpyrrolidone (PVP) is in the range from 50,000 to 400,000.
