Angiostatin fragments and aggregate angiostatin and methods of use

Abstract

Fragments and an aggregate form of an endothelial cell proliferation inhibit or and methods of use therefor are provided. The endothelial proliferation inhibitor is a protein derived from plasminogen, o r more specifically is an angiostatin fragment. The angiostatin fragments generally correspond to krinkle structures occurring within the endothial cell proliferation inhibitor. Angiostatin is also prepared in an aggregate form. The endothelial cell inhibiting activity of the angiostatin fragments and the aggregate angiostatin provide a means for inhibiting angiogenesis of tumors and for treating angiogenic-mediated disease.

    A pharmaceutical composition for use in inhibiting angiogenesis in a mammal comprising a pharmaceutically acceptable excipient and angiostatin fragment of plasminogen having angiogenesis inhibiting activity in a mammal, wherein the angiostatin fragment of plasminogen consists essentially of at least one kringle region of a plasminogen molecule and wherein the at least one kringle region does not correspond to an intact kringle 1 - 4 region.

    A composition for use in inhibiting angiogenesis in a mammal comprising a pharmaceutically acceptable carrier and an isolated DNA sequence that codes for an angiostatin fragment of plasminogen having angiogenesis inhibiting activity in a mammal wherein the angiostatin fragment of plasminogen consists essentially of one or more kringle region fragments selected from the group consisting of a kringle 1 region, a kringle 2 region, a kringle 3 region, a kringle 1 - 2 region, a kringle 1 - 3 region and a kringle 2-3 region, or a combination thereof.

    A compound for use in inhibition of angiogenesis in a mammal comprising a pharmaceutically acceptable carrier and a substantially purified aggregate angiostatin having angiogenesis inhibiting activity in a mammal, wherein the aggregate angiostatin is a recombinantly produced protein which precipitates in an approximately neutral pH aqueous solution and contains a kringle 1-4 region of a plasminogen molecule.

    The use of an angiostatin fragment of plasminogen in an administratable form for inhibiting endothelial cell proliferation, said angiostatin fragment of plasminogen having endothelial cell proliferation inhibiting activity in a bovine endothelial cell assay and wherein the angiostatin fragment of plasminogen consists essentially of one or more kringle region fragments of plasminogen selected from the group consisting of a kringle 1 region, a kringle 2 region, a kringle 3 region, a kringle 1-2 region and a kringle 2-3 region, or combination thereof.
