Human monoclonal antibodies to CTLA-4.

Abstract

In accordance with the present invention, there are provided fully human monoclonal antibodies against human cytotoxic T-lymphocyte antigen 4 (CTLA-4). Nucleotide sequences encoding and amino acid sequences comprising heavy and light chain immunoglobulin molecules, particularly contiguous heavy and light chain sequences spanning the complementarity determining regions (CDRS), specifically from within FR1 and/or CDR1 through CDR3 and/or within FR4, are provided. Further provided are antibodies having similar binding properties and antibodies (or other antagonists) having similar functionality as antibodies disclosed herein.

    AMENDED NATIONAL PHASE CLAIMS 1. A human monoclonal antibody that binds cytotoxic T lymphocyte antigen 4 (CTLA-4) or an antigen binding fragment thereof.

    5. The antibody or fragment according to any one of claims 1, 2 and 4, wherein said antibody inhibits human CTLA-4 binding to B7-1 with an IC 50 lower than about 0.50 nM and inhibits human CTLA-4 binding to B7-2 with an IC 50 lower than about 0.38 nM.

    8. The antibody or fragment according to any one of claims 1, 2 and 4 to 7 comprising a light chain CDR1, CDR2 or CDR3 sequence shown in any one of Figures 4 to 8.

    10. The antibody or fragment according to any one of claims 1, 2 and 4 to 9 comprising a light chain amino acid sequence selected from the group consisting of the amino acid sequences shown in SEQ ID NO:14-26, 65, 67, 69 or 71.

    11. The antibody or fragment according to any one of claims 1, 2 and 4 to 10, which has a heavy chain comprising the germline amino acid sequence of a human V H 333 (DP-50) gene or said germline sequence with at least 1 mutation.

    19. An antibody that competitively inhibits binding to CTLA-4 by antibody 4.1.1, 4.8.1, 6.1.1 or 11.2.1 having the amino acid sequence shown in Figure 22.

The antibody or fragment according to any one of claims 1, 2 and 4 to 5 comprising at least one of the following:a) human FR1, FR2 and FR3 sequences encoded by a human V H 3-33 ger family or said gene family with conservative substitutions, a single nonconservative substitution or both;or b) human FR1, FR2 and FR3 sequences encoded by a human Vk A27 ger family, or said gene family with conservative substitutions, a single non conservative substitution or both.

An antibody that binds to CTLA-4 that comprises an amino acid sequence sele from the group consisting of:a) an amino acid sequence comprising amino acid residues 1-89 of SEQ I NO: 5 operably linked to a CDR3 amino acid sequence that is encoded D7-27 gene and a JH4 gene;b) said amino acid sequence further comprising mutations at positions 24, and 47 of the amino acid residues 1-89 of SEQ ID NO: 5;and c) said amino acid sequence further comprising mutation’s at positions 1, and 7 of the CDR3 amino acid sequence.

    23. A cell culture system for assaying T cell stimulation, comprising a culture of human T cell blasts co-cultured with a Raji cell line.

    24. An assay for measuring T cell stimulation, comprising the steps of:a) providing a culture of human T cell blasts and a Raji cell line;b) contacting the culture with an agent;and c) measuring cytokine production by the culture.

    25. A pharmaceutical composition used in the treatment of cancer, inflammation or autoimmunity comprising the antibody or fragment according to any one of claims 1, 2 and 4 to 22 and a pharmaceutically acceptable carrier.
