Alteration of fcrn binding affinities or serum half-lives of antibodies by mutagenesis

Abstract

The present invention provides for a modified antibody of class IgG, in which at least one amino acid from the heavy chain constant region selected from the group consisting of amino acid residues 250, 314, and 428 is substituted with another amino acid which is different from that present in the unmodified antibody, thereby altering the binding affinity for FcRn and/or the serum half-life in comparison to the unmodified antibody.

    CLAIMS What is claimed is:1. A modified monoclonal antibody of class IgG with FcRn binding affinity altered relative to that of an unmodified monoclonal antibody of class IgG, comprising a heavy chain constant region wherein at least amino acid residues 250 and 428 are different from the residues present in the unmodified monoclonal antibody and wherein said unmodified monoclonal antibody is selected from the group consisting of an anti-CD25, an anti-CD3, an anti-IFNγ, and an anti-α5βl integrin.

    An anti-CD25 antibody of class IgG comprising a heavy chain amino acid sequence at least 95% identical to a sequence selected from the group consisting of SEQ ID NOs:122, 123, 127, and 128, and wherein residue 250 is glutamine and residue 428 is leucine or phenylalanine.

    An anti-CD3 antibody of class IgG comprising a heavy chain amino acid sequence at least 95% identical to a sequence selected from the group consisting of SEQ ID NOs:139, and 140, and wherein residue 250 is glutamine and residue 428 is leucine or phenylalanine.

    An anti-α5βl integrin antibody of class IgG comprising a heavy chain amino acid sequence at least 95% identical to a sequence selected from the group consisting of SEQ ID NOs:145, and 146, and wherein residue 250 is glutamine and residue 428 is leucine or phenylalanine.

    An anti-IFNγ antibody of class IgG comprising a heavy chain amino acid sequence at least 95% identical to a sequence selected from the group consisting of SEQ ID NOs:133, and 134, and wherein residue 250 is glutamine and residue 428 is leucine or phenylalanine.
