  1. 1
A method of determining whether a substance is an inhibitor or an activator of a protein whose presence in a cell evokes a responsive change in a phenotypic characteristic other than the level of said protein in said cell per se, which comprises:a) providing at least.one cell containing said protein, b) treating said cell containing said protein with said substance, and c) examining the treated cell to determine whether it exhibits a change in a phenotypic characteristic in response to said substance.
  2. 2
, A method of determining whether a substance is an inhibitor or activator of a protein whose production 5 by a cell evokes a responsive change in a phenotypic characteristic other than the level of said protein in said cell per se, which comprises:« (a) providing a first cell line which 10 * produces said protein and exhibits said phenotypic response to the protein;(b) providing a. second cell line which produces the protein at a lower level than 15 the first cell line, or does not produces the protein at all, and which exhibits said phenotypic response to the protein to. ..a lesser degree or not at all;20 (c) incubating the substance with the first and second cell lines;and ,(d) comparing the phenotypic response of the first cell line to the substance with the 25 phenotypic response of the second cell line ., to the substance. '
  3. 22
a test kit for determining whether a substance is an inhibitor or activator of a protein whose production evokes a responsive change ,in a phenotypic characteristic other than level of said protein in said 5 cell per se, according to the method of Claim 2., which comprises:(a) a first cell line which produces the protein and exhibits said phenotypic response thereto;and 10 .(b) a second cell line which produces the protein at a lower level than the first cell line, or does not produce the protein, and which exhibits said phenotypic response to the protein to a lesser degree or not at all.
  4. 25
25 agar, colony size *in soft agar, and combinations thereof. 25. The method of claim 2 wherein the response is a change in an antigenic characteristic of the cell. PA 89227/2
  5. 26
A method of determining whether a substance is an inhibitor or an activator of a protein whose production by a cell evokes a responsive change in a phenotypic characteristic other than the level of said protein in said cell per se, which comprises:a) providing a cell which has been genetically altered to overproduce a selected protein, b) treating said cell containing the overproduced selected protein with said substance, and c) examining the treated cell to determine whether it e-rMT-KEg a change in a phenotypic characteristic in response to said substance.
  6. 27
A method of determining whether a substance is an inhibitor or an activator of a protein whose production by a cell evokes a responsive change in a phenotypic characteristic other than the level of said protein in said cell per se, according to. claim26 ־ ; which comprises:¢1) providing a cell which has been genetically altered to overproduce a selected protein, ־ b) treating said cell containing the overproduced selected protein with said substance, and PA 89227/2 c). examining the treated cell to determine whether it exhibits a graded cellular response to said substance.
  7. 31
The method according to claims 26 or 27. wherein the genetically altered cell is selected from the group consisting of cells which have been genetically engineered by insertion of a nucleic acid sequence and cells which have been mutated to increase expression of the selected protein.
  8. 32
32? a method of determining whether a substance is an inhibitor ׳. or an activator of a protein whose production by a cell evokes a PA 89227/ 3 responsive change in a phenotypic characteristic other than the level of said protein in said cell per se, which comprises :נ a) providing a first cell which produces said protein and exhibits said phenotypic characteristic in response to the protein;b) incubating the substance with the first cell, and c) coinparing the phenotypic response of the first cell in the presence of the substance with the phenotypic response of a second cell in the absence of the substance.
  9. 35
The methods of claims 26 or 27 wherein the examination includes comparing the phenotypic response of the first cell in the presence of the substance with the phenotypic response of the second cell in the presence of a known inhibitor or activator of the protein. PA 89227/ 3
  10. 39
. The method of, claim 26 wherein said cell is obtained by introducing a gene encoding the protein of interest into a host cell, said gene being under the control of a promoter functional in the’ host cell, whereby said gene is expressed.
  11. 45
A cell into which a selected protein has been introduced, said protein having been found capable of evoking a change in a phenotypic characteristic of said cell in response to a selected substance which has been found to be an inhibitor .or activator of said protein by the method of claims 26, 27 or 32.
