Methods, compositions, and devices for supplying dietary fatty acid needs

Abstract

Nutritional formulas comprising long-chain polyunsaturated fatty acids (LC-PUFAs) are provided, along with methods and devices for preparing and/or administering nutritional formulas. In some embodiments, a percentage of the LC-PUFAs in the nutritional formula are in the form of monoglycerides and/or free fatty acids. In some embodiments, the nutritional formulas do not comprise added lipase. Also provided are methods for providing nutrition to a subject, methods for improving fat absorption, methods for improving cognitive ability, methods for preventing chronic lung disease, and methods for reducing the length of time a patient requires total parenteral nutrition.

    A device for providing a nutritional formula comprising monoglycerides and free fatty acids by temporarily exposing a nutritional composition comprising long-chain polyunsaturated fatty acid (LC-PUFA) triglycerides and/or LC-PUFA esters to lipase prior to ingestion, the device comprising:a container defining an interior for receiving the nutritional composition;a structure in fluid communication with the interior of the container;and at least one lipase immobilized to the structure so that the nutritional composition is exposed to the at least one lipase when the nutritional composition is received within the container, wherein the device is a feeding mechanism or includes an opening configured to connect to a feeding mechanism.

    The device of any one of claims 1-3, wherein the feeding mechanism is at least one of a bottle or a nasogastric tube.

    The device of any one of claims 1-4, wherein the at least one lipase is selected from the group consisting of Chromobacterium viscosum lipase, Pseudomonas fluorescens lipase, and Rhizopus oryzae lipase.

    The device of any one of claims 1-5, wherein the container is configured so that when the nutritional formula exits the container, the at least one lipase remains in the container.

    The device of any one of claims 1-7, wherein the lipase is immobilized by at least one of covalent binding, adsorption, ionic binding, cross-linking, encapsulation, or entrapment.

    The device of any one of claims 1-8, wherein the lipase is immobilized within the container such that when the nutritional formula exits the container, the lipase remains in the container.

    A method of preparing a nutritional formula comprising LC-PUFAs in the form of monoglycerides and free fatty acids prior to ingestion by a subject, comprising:exposing a liquid nutritional composition comprising LC-PUFA triglycerides and/or LC-PUFA esters to an immobilized lipase;and separating the immobilized lipase from the liquid nutritional composition after exposing the LC-PUFA triglycerides and/or LC-PUFA esters to the immobilized lipase, wherein the nutritional formula is ready for ingestion by the subject once the liquid nutritional composition is exposed to the immobilized lipase and the liquid nutritional composition is separated from the immobilized lipase.

    The method of claims 10 or 11, wherein the lipase is immobilized to a solid support, and the immobilized lipase is removed by separating the liquid nutritional composition from the solid support.

    The method of any one of claims 10-12, comprising passing the liquid nutritional composition through a container to expose the liquid nutritional composition to the immobilized lipase, wherein the lipase is immobilized to particles within the container.

    The method of any one of claims 10-13, comprising passing the liquid nutritional composition through a container comprising lipase immobilized to a solid support to expose the liquid nutritional composition to the immobilized lipase, wherein the liquid exits the container by passing through a material that is permeable to the liquid nutritional composition but not permeable to the solid support to separate the immobilized lipase from the liquid nutritional composition, and wherein optionally the liquid nutritional composition exits the container through a feeding tube.
