Abstract

The present invention relates to HCV variants, particularly variants that are resistant to a protease inhibitors such as VX-950. Also provided are methods and compositions related to the HCV variants. Further provided are methods of isolating, identifying, and characterizing multiple viral variants from a patient.

    An isolated HCV polynucleotide encoding an HCV NS3 protease, an analog thereof, or a fragment thereof;wherein the protease, the analog thereof or the fragment thereof has serine protease activity;and, wherein an amino acid selected from the group consisting of lysine, threonine, serine, isoleucine, methionine, leucine and glycine is encoded at the position that corresponds to position 155 of a polynucleotide sequence encoding a wildtype HCV NS3 protease.

    An HCV variant comprising a polynucleotide according to any of claims 1 or 2.

    An isolated HCV protease comprising an amino acid sequence in which the amino acid at the position corresponding to amino acid position 155 of a wildtype HCV NS3 protease amino acid sequence is an amino acid selected from the group consisting of lysine, threonine, serine, isoleucine, methionine, leucine and glycine.

    An expression system comprising the HCV polynucleotide of any one of claims 1 or 2.
