Improved grain quality through altered expression of seed proteins

Abstract

The present invention is directed to compositions and methods for altering the levels of seed protein in cereal grain. The invention is directed to the alteration of seed protein levels in plant grain, resulting in grain with increased digestibility/nutrient availability, improved amino acid composition/nutritional value, increased response to feed processing, improved silage quality, and increased efficiency of wet milling.

    A genetically modified cereal plant having improved grain quality compared to the corresponding unmodified plant;the improved grain quality being due to a stably decreased level of a zein protein in the grain, or to a stably increased level of an alpha-globulin protein or a legumin1 protein in the grain.

    A plant according to any one of the preceding claims, wherein said decreased level of said zein is due to the stable introduction of means for transposon-tagging, co-suppression, antisense suppression or RNAi-mediated suppression;or to a mutation, and preferably wherein said decreased level of said zein is due to a mechanism other than antisense expression.

    A plant according to any one of the preceding claims, wherein said decrease in the level of said zein is at least about 5%, 10%, 20%, 50%, 70%, 90%, 95% or 99%, or wherein said increase in the level of said alpha-globulin or legumin1 is at least about 5%, 10%, 20%, 50%, 70%, 90%, 100%, 200% or 500%.

    A plant according to any one of the preceding claims which is a maize, wheat, barley, rice, rye, oat or sorghum plant.

    A plant according to any one of the preceding claims wherein the coding sequence of the zein whose level is reduced is the sequence of the 50KD gamma-zein of SEQ ID NO:1 or a sequence with at least 80%, 90%, 95% or 99% sequence identity thereto.

    A plant according to any one of the preceding claims wherein the improvement in grain quality is the result of at least one of:(a) increased energy availability when the grain is eaten;(b) increased digestibility;(c) decreased anti-nutritional properties of the grain;(d) improved properties in wet-milling;and (e) reduced nitrogen excretion into the environment by animals that have eaten the grain.

    A plant according to any one of the preceding claims wherein the handling properties of the grain are unaltered, or substantially unaltered.

    A plant according to any one of the preceding clams wherein the grain retains a vitreous phenotype.

    A method of making a cereal plant with improved grain quality according to any one of claims 1 to 12, comprising stably decreasing by genetic modification of a cereal plant cell, or by mutation, the level of said zein or stably increasing by genetic modification of a cereal plant cell said alpha-globulin or legumin1, and preferably wherein said stable decrease is obtained by the stable introduction of means for transposon-tagging, co-suppression, antisense suppression or RNAi-mediated suppression, or by mutation;or said stable increase is achieved by the introduction of a sequence encoding an alpha-globulin or legumin1 protein.

    A method according to 14 further comprising regenerating a genetically modified plant from said cell, and optionally:(a) selecting for a genetically modified plant with improved grain quality;(b) and/or obtaining genetically modified progeny plants of one or more further generations from said plant.

    A method according to any one of claims 14 to 16 further comprising obtaining seed with a decreased level of said zein or an increased level of said alpha-globulin or legumin1 from said plant.

    A method according to any one of claims 14 to 17 further comprising obtaining said grain having said improved quality, and optionally processing said grain and/or feeding said grain to animals.

    A genetically modified plant cell, plant part, plant, progeny plant or feed, or grain with improved grain quality;obtained or obtainable by the method of any one of claims 14 to 18.

    Use of a nucleotide sequence which suppresses gene expression to improve grain quality in a cereal plant by stably decreasing the level of a zein in the grain;or of a sequence encoding an alpha-globulin or legumin1 protein to improve grain quality in a cereal plant by stably increasing the level of an alpha-globulin or legumin1 in the grain.
