Method of administering therapeutic polypeptides, and polypeptides therefor

Abstract

The invention relates to a method suitable for administering protein therapeutic molecules orally, sublingually, topically, intravenously, subcutaneously, nasally, vaginally, rectally or by inhalation so as to avoid inactivation, by using VHH polypeptides derived from Camelidae antibodies. The invention further relates to the said therapeutic molecules. The invention further relates to a method for delivering therapeutic molecules to the interior of cells. The invention further relates to anti-IgE therapeutic molecules.

  1. 1
A polypeptide construct comprising at least one single domain antibody directed against an internalising cellular receptor, and at least one therapeutic polypeptide or agent or an antigen.
  2. 2
A polypeptide construct comprising at least one single domain antibody directed against an internalising cellular receptor, and at least one single domain antibody directed against a therapeutic target.
  3. 3
A polypeptide construct according to claims 1 or 2, wherein said internalising cellular receptor is Epidermal Growth Factor receptor.
  4. 5
A polypeptide construct according to claims 1 or 2, wherein said internalising cellular receptor is any of LDL receptor, FGF2r, ErbB2r, transferring receptor, PDGr, VEGFr, PsmAr or antigen presenting cell internalising receptor.
  5. 6
A polypeptide construct according to claims 1 or 2, wherein the single domain antibody directed against an internalising cellular receptor specifically binds to a receptor on the intestinal wall which induces an active transfer through the wall.
  6. 7
A polypeptide construct according to any of claims 1 to 5, wherein the construct comprises a drug or toxic compound.
  7. 8
A polypeptide construct according to any of claims 1 to 5, wherein the single domain antibody directed against a therapeutic target is directed against PDK1.
  8. 9
A polypeptide construct according to any of claims 1 to 5, wherein the single domain antibody directed against a therapeutic target is directed against any of GSK1, Bad, caspase and Forkhead.
  9. 10
Use of a polypeptide construct according to any of claims 8 or 9 for the preparation of a medicament for treating cancer.
  10. 11
A polypeptide construct according to any of claims 1 to 5, comprising a single domain antibody directed against an internalising receptor on an antigen presenting cell (APC) and further comprising an antigen.
  11. 12
Method for delivering a therapeutic polypeptide, agent or antigen to the interior of a cell comprising covalently or non-covalently attaching thereto a polypeptide construct comprising at least one single domain antibody directed against an internalising cellular receptor.
  12. 13
Method for delivering a therapeutic polypeptide and/or agent or antigen to the interior of a cell without being inactivated comprising covalently or non-covalently attaching thereto a single domain antibody directed against an internalising cellular receptor.
  13. 14
Method for delivering a therapeutic polypeptide and/or agent or antigen across a natural barrier comprising covalently or non-covalently attaching thereto a single domain antibody directed against an internalising cellular receptor.
  14. 16
Method for delivering an antigen to the inside of an antigen presenting cell comprising linking said antigen to a single domain antibody specific for an internalising cellular receptor on an antigen presenting cell.
  15. 21
A nucleic acid encoding a polypeptide construct according to any of claims 1 to 9, 11 or 17 to 20.
  16. 22
A composition comprising a polypeptide construct as defined in any of claims 1 to 9, or 17 to 20, together with a pharmaceutical carrier.
