Vascular endothelial cell growth factor antagonists and uses thereof

Abstract

The present invention provides vascular endothelial cell growth factor (VEGF) antagonists and methods of using VEGF antagonists. VEGF antagonists contemplated by the invention include VEGF antibodies and VEGF receptor fusion proteins. Methods of treating edema and stroke using VEGF antagonists are also provided.

    Use of an hVEGF antagonist in the manufacture of a medicament for treating a mammal having edema associated with a neoplastic disease or condition wherein the neoplastic disease or condition is breast carcinoma, lung carcinoma, gastric carcinoma, esophageal carcinoma, colorectal carcinoma, liver carcinoma, ovarian carcinoma, thecoma, arrhenoblastoma, cervical carcinoma, endometrial carcinoma, endometrial hyperplasia, endometriosis, fibrosarcoma, choriocarcinoma, neck cancer, nasopharyngeal carcinoma, laryngeal carcinoma, hepatoblastoma, Kaposi's sarcoma, melanoma, skin carcinoma, hemangioma, cavernous hemangioma, hemangioblastoma, pancreas carcinoma, retinoblastoma, rhabdomysarcoma, osteogenic sarcoma, leiomyosarcoma, urinary tract carcinoma, thyroid carcinoma, Wilm's tumor, renal cell carcinoma, prostate carcinoma, phakomatosis, or Meigs' syndrome.

    Use of an hVEGF antagonist in the manufacture of a medicament for preventing, reducing, or inhibiting edema in a mammal associated with a neoplastic disease or condition, wherein the neoplastic disease or condition is breast carcinoma, lung carcinoma, gastric carcinoma, esophageal carcinoma, colorectal carcinoma, liver carcinoma, ovarian carcinoma, thecoma, arrhenoblastoma, cervical carcinoma, endometrial carcinoma, endometrial hyperplasia, endometriosis, fibrosarcoma, choriocarcinoma, neck cancer, nasopharyngeal carcinoma, laryngeal carcinoma, hepatoblastoma, Kaposi's sarcoma, melanoma, skin carcinoma, hemangioma, cavernous hemangioma, hemangioblastoma, pancreas carcinoma, retinoblastoma, rhabdomysarcoma, osteogenic sarcoma, leiomyosarcoma, urinary tract carcinoma, thyroid carcinoma, Wilm's tumor, renal cell carcinoma, prostate carcinoma, phakomatosis, or Meigs' syndrome.

    Use according to any one of Claims 1-2, wherein the neoplastic condition comprises a solid tumor.

    Use according to any one of Claims 1-3, wherein the mammal is a human.

    Use according to any one of Claims 1-4, wherein the hVEGF antagonist comprises an anti-hVEGF antibody.

    Use according to any one of Claims 5-7, wherein said antibody comprises a monoclonal antibody.

    Use according to any one of Claims 1-4, wherein the hVEGF antagonist comprises an hVEGF receptor fusion protein.

    Use according to any one of Claims 1-11, wherein the hVEGF antagonist is administered serially or in combination with chemotherapy or radiation.

    Use according to any one of Claims 1-12, wherein the hVEGF antagonist is administered serially or in combination with tumor necrosis factor, alpha-interferon, beta-interferon, gamma-interferon, anti-HER2 antibody, heregulin, anti-heregulin antibody, D-factor, interleukin-1, interleukin-2, or granulocyte macrophage stimulating factor.

    Use according to any one of Claims 1-14, wherein the hVEGF antagonist is administered serially or in combination with an agent that promotes microvascular coagulation in tumors.
