Glycoprotein synthesis

Abstract

Methods for making glycoproteins, both in vitro and in vivo, are provided. One method involves incorporating an unnatural amino acid into a protein and attaching one or more saccharide moieties to the unnatural amino acid. Another method involves incorporating an unnatural amino acid that includes a saccharide moiety into a protein. Proteins made by both methods can be further modified with additional sugars.

    Claims of equivalent WO 2004035605 A2 CLAIMS WHAT IS CLAIMED IS:1. A method for synthesis of a glycoprotein, the method comprising: a) incorporating into a protein an unnatural amino acid that comprises a first reactive group;and, b) contacting the protein with a saccharide moiety that comprises a second reactive group, wherein the first reactive group reacts with the second reactive group to attach the saccharide moiety to the unnatural amino acid.

    A composition comprising a translation system, the translation system comprising an orthogonal tRNA (O-tRNA) and an orthogonal aminoacyl tRNA synthetase (O-RS), wherein the O-RS preferentially aminoacylates the O-tRNA with an unnatural amino acid that comprises a saccharide moiety and the O-tRNA recognizes at least one selector codon.
