Monoclonal antibody against flt4 receptor tyrosine kinase and its use in diagnosis and therapy

Abstract

Anti-FLT4 antibodies, especially monoclonal anti-FLT4 antibodies, which are useful as a specific marker for endothelial cells of lymphatic vessels and HEVs, as a diagnostic tool for detecting changes in lymphatic tissue, especially in lymphatic vessels and HEVs in disease states, such as lymphangioma, metastatic lymph nodes and inflammatory, infectious and immunological disease.

7 claims: 6 independent, 1 dependent

  1. 1
Use of a monoclonal antibody directed against the extracellular domain of the FLT4-receptor tyrosine kinase for the preparation of a diagnostic composition for imaging lymphatic vessels, lymph nodes or high endothelial venules (HEVs).
  2. 2
Use of a monoclonal antibody directed against the extracellular domain of the FLT4-receptor tyrosine kinase for the preparation of a diagnostic composition for detecting lymphatic tissue, lymphatic vessels or high endothelial venules (HEVs).
  3. 4
Use of a monoclonal antibody directed against the extracellular domain of the FLT4-receptor tyrosine kinase for the preparation of pharmaceutical composition for inhibiting the FLT-4 mediated lymphatic vascularization which is associated with a disease selected from the group consisting:metastatic cancers, lymphomas, lymphangiomas, inflammation (chronic or acute), infections and immunological diseases.
  4. 5
The use of any one of claims 1 to 4, wherein said monoclonal antibody is an anti-FLT4 monoclonal antibody produced by a hybridoma cell line deposited as DSM ACC 2210.
  5. 6
A method for in vitro imaging lymphatic vessels in a tissue sample, comprising the steps of:(a) applying a detectably labeled anti-FLT4 antibody to said tissue sample suspected of containing lymphatic vessels;and (b) detecting said detectably-labeled anti-FLT4 antibody bound to said tissue sample.
  6. 7
A method for diagnosing diseases characterized by changes in lymphatic vessels and HEVs, comprising the steps of:(a) exposing a tissue sample obtained from a patient suspected of having a disease characterized by changes in lymphatic cells and HEVs to a labeled anti-FLT-4 antibody;(b) washing said tissue sample;and (c) detecting the presence of said detectably-labeled anti-FLT-4 antibody in said tissue sample.
