Tissue-preferential promoters.

Abstract

DNA sequences are able to function as promoters of tissue-preferential transcription of associated DNA sequences in plants, particularly in the roots. These DNA sequences can be used in transformation vectors to produce transgenic plants which will express the heterologous genes preferentially in tissue, particularly in the roots of maize plants..

    A process for the production of an plant expression vector containing a tissue-preferential promoter DNA oparably linked to a DNA expressible in plants which process comprises (a) inserting said tissue-preferential promoter DNA in front of the DNA expressible in plants;and(b) splicing this construct in correct reading frame into a known plant expression vector which may contain one or more marker genes suitable for transformant selection. A recombinant DNA sequence comprising a tissue-preferential promoter DNA sequence obtainable from a maize plant.

    A recombinant DNA sequence comprising a tissue-preferential promoter DNA sequence having substantial sequence homology to the DNA sequence of Figure 9. The process according to claim 5, wherein the tissue-specific promoter DNA sequence is obtainable from a maize inbred plant.

    A recombinant DNA sequence comprising a tissue- preferential promoter DNA sequence which is obtainable from a naturally occuring DNA sequence encoding a metallothionein-like protein. The process according to claim 6, wherein the tissue-specific promoter DNA sequence is obtainable from the maize inbred line Funk 211D.

    The process according to any one of claims 1 to 3, wherein the tissue-preferential promoter DNA sequence has substantial sequence homology to the DNA sequence of Figure 9. The recombinant DNA sequence according to any one of claims 1,6 and 7, wherein said tissue-preferential promoter DNA sequence is operably linked to a DNA coding sequence.
