Cross-linked glucose isomerase.

Abstract

The invention relates to a novel water-insoluble glu­cose isomerase which is formed by a crystalline enzyme converted to solid form by cross-linking. The inven­tion also concerns a process for the preparation of the novel crystalline glucose isomerase by cross-link­ing with dialdehyde in the presence of a compound con­taining at least one amino group, and the use of this novel enzyme preparation as an isomerization catalyst.

    A crystalline glucose isomerase, charac­terized in that a crystalline soluble glucose isomerase is converted to water-insoluble form by cross-linking.

    A glucose isomerase according to the claims 1 to 5, characterized in that the crystals are chemically or physically bound into larger bodies, spherical, sheetlike, bandlike or the like combinations.

    A reactor useful for the continuous production of fructose, said reactor containing water-insoluble substantially pure, cross-linked crystalline glucose isomerase.

    A process for the preparation of a water-­insoluble cross-linked crystalline enzyme, charac­terized by a) adding a dialdehyde to a crystalline isomerase suspension wherein, before the addition of dialdehyde or during the cross-linking reaction a compound contain­ing at least one amino group, such as ammonium salt, amine or amino acid is added thereto, and b) washing the cross-linked crystals by water or some other liquid for removing reagent residues.
