Amplifying the expression of recombinant DNA products.

Abstract

Amplified expression of recombinant DNA products is achieved in hosts expressing protease that cleave at multi-basic amino acid residues. To this end, wild-type genes encoding the desired protein products are mutated by substituting codons or eliminating codons encoding multi-basic amino acid residues while maintaining the activity of the expressed protein product. Mutation of the desired gene can be conveniently carried out by site-specific in vitro mutagenisis.

    A method as claimed in any one of Claims 11, 12 and 13, wherein the recombinant DNA encodes analog granulocyte-macrophage colony stimulating factor, with at least one basic amino acid residue in the naturally occurring granulocyte-mactrophage colony stimulating factor being:replaced with a non-basic amino acid residue to eliminate multibasic amino acid residues;and/or,deleted to eliminate multibasic amino acid residues.

    A method as claimed in any one of Claims 9, 10 and 11, further comprising the step of preparing recombinant DNA encoding a polypeptide devoid of multibasic amino acid residues by:replacing at least one codon encoding a basic amino acid residue in the DNA encoding the naturally" occurring form of the polypeptide with a codon encoding a non-basic amino acid residue;or,deleting from the DNA encoding the naturally occurring form of the polypeptide at least one codon encoding a basic amino acid residue.

    A method as claimed in any one of Claims 11, 12, 13 and 15, wherein the recombinant DNA encodes analog granulocyte-macrophage colony stimulating factor.
