Fc.gamma.riib-specific antibodies and methods of use thereof

Abstract

The present invention relates to antibodies or fragments thereof that specifically bind the extracellular domain of Fc.gamma.RIIB, particularly human Fc.gamma.RIIB, and block the Fc binding site of human Fc.gamma.RIIB. The invention provides methods of treating cancer and/or regulating immune complex mediated cell activation by administering the antibodies of the invention to enhance an immune response. The invention also provides methods of breaking tolerance to an antigen by administering an antigen-antibody complex and an antibody of the invention.

Use of a therapeutically effective amount of an antibody or fragment thereof in combination with an antigen-antibody complex comprising an antigen for the manufacture of a medicament for breaking tolerance to said antigen in a patient, wherein said antibody or said fragment thereof comprises a variable domain that specifically binds the extracellular domain of human FcyRIIB that is endogenously expressed on the surface of a cell and blocks the Fc binding site of said human FcyRIIB.

    A pharmaceutical composition for breaking tolerance to an antigen comprising (i) a therapeutically effective amount of an antibody or fragment thereof comprising a variable domain that specifically binds the extracellular domain of human FcyRIIB that is endogenously expressed on the surface of a cell, and blocks the Fc binding - 143 CA 02590935 2013-10-11 site of human FcyRIIB;(ii) an antigen-antibody complex comprising said antigen;and (iii) a pharmaceutically acceptable carrier.
