Compositions of restricted cells capable of rapid growth which produce proliferation suppressive materials, and uses thereof

Abstract

Compositions of matter are described which contain restricted proliferative cells. When so restricted, the cells produce an unexpectedly high amount of material which suppresses cell proliferation. The phenomenon crosses cell type and species lines. Processes for making these compositions, and their use, are also described.

    CA 02334483 2008-03-27 CLAIMS:1. Composition useful in suppressing proliferation of cells, produced by entrapping a sample of rapidly proliferating cells in a biocompatible, proliferation-restrictive, selectively-permeable agarose bead coated with agarose or agarose and collagen bead coated with agarose, culturing said bead in culture medium for a time sufficient to restrict proliferation of said entrapped cells, wherein said entrapped cells produce a cell proliferation suppressing material inside said bead which suppresses proliferation of the cells and filtering the medium through a filter which separates material having a molecular weight of at least about 30 kd from material having a molecular weight of less than 30 kd and recovering the composition comprising the material having a molecular weight of at least about 30 kd.

    The composition of any one of claims 1 to 5, wherein said entrapped cells are human cells.

    A process for stimulating production of a material which has a cell proliferationinhibiting effect, comprising culturing a biocompatible, proliferation-restrictive, selectively-permeable agarose bead coated with agarose or agarose and collagen bead coated with agarose which has rapidly proliferating cells entrapped therein in a medium for a time sufficient for said cells to produce proliferation-suppressing material inside said bead, and filtering the medium through a filter which separates material having a molecular weight of at least about 30 kd from material having a molecular weight of less than 30 kd, and recovering the material having a molecular weight of at least about 30 kd.

    The process of any one of claims 10 to 14, wherein said medium is serum free.

    A method for increasing production of a material that suppresses cell growth and has a molecular weight of at least 30 kd, comprising restricting highly proliferative cells with a bead-forming material to form a biocompatible, selectively-permeable, proliferation-restrictive proliferative cell containing agarose bead coated with agarose or agarose and collagen bead coated with agarose and culturing the cells until they are restricted and produce more of said at least 30 kd material inside said bead than an equal number of unrestricted cells, and then filtering through a filter which separates material having a molecular weight of at least about 30 kd from material having a molecular weight of less than 30 kd to yield said material having a molecular weight of at least about 30 kd.
