The cytoplasmic inhibition of gene expression

Abstract

One aspect of the invention is to provide novel geneticconstructions for the expression of inhibitory RNA in thecytoplasm of eukaryotic cells. The inhibitory RNA may bean anti-sense RNA or a co-suppressor RNA. The geneticconstructions of the invention are capable of replicating in thecytoplasm of a eukaryotic cell and comprise a promoter regionin functional combination with an encoding polynucleotide. The genetic constructions may be designed so as to replicate inthe cytoplasm of plant cells, yeast cells, and mammalian cells. When the eukaryotic cell of interest is a plant cell, the geneticconstruction is preferably derived from a plant RNA virus. Plant RNA virus derived generic constructions , may employa plant virus subgenomic promoter, including subgenomicpromoters from tobamoviruses in functional combination withthe RNA encoding region. Another aspect of the inventionis to provide cells comprising the genetic constructions ofthe invention and organism comprising a plurality of suchcells. Another aspect of the invention is to provide methodsof reducing the expression of a gene of interest in eukaryoticcells, i.e., methods of producing eukaryotic cells exhibitingreduced levels of expression of a gene of interest. The methodsof the invention comprise the step of transfecting a cell witha genetic construction of the invention in which the RNAencoding region is specific for the gene of interest. Anotheraspect of the invention is to provide plant cells that produceelevated levels of the carotenoid phytoene. The elevated levelsof phytoene are achieved by inhibiting the expression at theenzyme phytoene desaturase using the vectors of the invention.

A method of reducing expression of a gene of interest in a plant, the method comprising transfecting one or more plant cells of the plant with a genetic vector comprising: (a) a first plant viral subgenomic promoter operably joined to a first nucleic acid sequence that codes for a plant viral coat protein wherein transcription of the first nucleic acid sequence is regulated by the first plant viral subgenomic promoter: (b) a second plant viral subgenomic promoter operably joined to a second nucleic acid sequence which codes for an anti-sense RNA or a co-suppressor RNA specific for a gene of interest in a plant wherein transcription of the second nucleic acid sequence is regulated by the second plant viral subgenomic promoter;and, (c) an origin of replication that initiates replication of the genetic vector in the cytoplasm of the plant cell;wherein the nucleic acid sequence which codes for an antisense RNA or a cosuppressor RNA is specific for the gene of interest.

    A method of producing a plant cell having reduced expression of a phytoene desaturase, the method comprising transfecting the plant cell with a genetic vector comprising:(a) a first plant viral subgenomic promoter operably joined to a first nucleic acid sequence that codes for a plant viral coat protein wherein the transcription of the first nucleic acid sequence is regulated by the first plant viral subgenomic promoter;(b) a second plant viral subgenomic promoter operably joined to a second nucleic acid sequence which codes for an anti-sense RNA or a co-suppressor RNA specific for phytoene desaturase wherein transcription of the second nucleic acid sequence is regulated by the second plant viral subgenomic promoter;and (c) an origin of replication that initiates replication of the genetic vector in the cyto plasm of a plant cell: wherein the nucleic acid sequence which codes for an antisense RNA or a cosuppressor RNA is specific for phytoene desaturase.
