Abstract

one. The method of determining the status of ploidy of a chromosome or chromosome segment in a fetus involving: measuring DNA in a sample which contains maternal DNA from the mother of the fetus and fetal DNA from the fetus, in the set of polymorphic loci in the chromosome or chromosome segment; computer calculation of the number of alleles in the set of polymorphic loci from DNA measurements, made in the sample; creating on the computer a variety of hypotheses of ploidy, each of which refers to a different possible state of chromosome ploidy or chromosome segment; building a joint distribution model for the expected number of alleles in a variety of polymorphic loci in a chromosome or chromosome segment for each ploidy hypothesis; determining the relative probability of each ploidy hypothesis using a joint model distributions and numbers of alleles, measured in the sample; and determining the state of fetal ploidy by selecting the state of ploidy, corresponding to the hypothesis with the highest probability. The method of claim 1, wherein the DNA in the sample is derived from the maternal plasma. The method of claim 1, wherein the method further provides for preferential multiplication of DNA in the sample at a plurality of polymorphic loci prior to the measurement step.
