The appearance of the tumor was followed by weekly examination of the animals. About 90% of the animals developed tumors between 35 and 70 days after induction.
The tumor weight was determined using the method of H. Druckrey, D. Steinhoff, M. Nakayama, R. Preussmann, K. Anger (1983) Experimentelle Beitrage zum Dosis-Problem in der Krebs Chemotherapie Żur Wirkungweise von Endoxan, Dtsch. Med. Wschr. 88: 651. The method involved comparing tumor weights determined by hand examination and determined by direct weighing (after tumor resection).
The correlation coefficient was 0.98. After reaching a total tumor weight of about 1 g, the animals were randomized and 7 were placed in control and study groups.
Treatment was started immediately after subcutaneous injection of the test substance.
The hormone status in animals was determined by methylene blue vaginal staining of the cell smear and evaluation by the method of TC Jones, U. Mohr, RD Hunt (1972); The genital system, in: Monographs on pathology of laboratory animals sponsored by the International Life Science Institute (Springer, NY, London).
The results of the experiments are presented on flg. 1. The tumor weight curve for animals in the untreated control group showed unrestrained growth. Curves 1 (*) and 2 (0) show cetrorelix acetate treatment in two different carriers. The extended curve 3 shows a drastic reduction in tumor weight after pamoate treatment.
Since the treatment in this case involved only one dose treatment, the tumor increased because treatment with one dose would not kill all cancer cells.
The agent according to the invention is an amorphous precipitate, as shown by X-rays of the decapeptide-cetrorelix in the form of the embonic acid salt. The aqueous suspension of this precipitate, which may optionally contain isotonic additives, clearly showed a prolonged effect on the animal model compared to the aqueous solution of pep6
186 929 weeks It was surprisingly found that the duration of action was the same as the time for the injection suspension that contained peptide pamoate pellet in a biodegradable polymer such as lactic and glycolic acid copolymers. This result was particularly surprising because, as outlined above, until now only long-acting medical forms produced in a very labor-intensive manner that contained the active substance in biologically degradable polymers had sufficiently long duration of action.
This was also unexpected because, according to J. Pharm. Pharmavol. 47,878-883 (1985), pyrimethamine, a 2,4-diaminopyrimidine derivative, showed no difference compared to its pamoic acid salt in terms of pharmacokinetic behavior, plasma levels and AUC after subcutaneous injection in mice.
Similarly, imipramine hydrochloride showed no difference when compared to oral imipramine pamoate (Indian Journal of Physiology and Pharmacology, 25 94), 331-338 (1989). Suspensions for injection of crystalline non-peptide medicinal substances such as prednisolone or triamcinolone are known in the prolonged form as in the case of the insulin zinc salt suspension used to treat diabetes. Insulin consists of 51 amino acids. All recently mentioned forms are crystalline, while X-ray diffraction analysis has shown that the form according to the invention is amorphous. The particle size of the agent according to the invention is in the range of 5 μη - 200 pm. Ce1oorelix emoonate with particles less than 5 μηι produced a more pronounced wild effect than the agent of the invention. Similarly, emetico cetrorelix with a particle size greater than 200 m has a weaker effect of prolonged action than the agent of the invention.
While the advantages of the agent according to the invention consist in better reproducibility of results, the quality of the drug produced with the agent according to the invention is therefore less affected.
Example 1
At an equimolar ratio of peptide (calculated relative to the free base) to embonic acid, an aqueous solution of embonic acid in excess is combined with a solution of cetrorelix acetate to precipitate embonic acid in the form of yellow crystals. After addition of dilute sodium hydroxide solution to pH 7-7.5, pamoic acid dissolved and precipitated from the decapeptide as the salt of cetrorelix pamoate with a molar ratio of peptide: pamoic acid 2: 1 (mol / mol). The precipitate was filtered off, washed with water and dried.
Example 2
Cetrorelix acetate and pamoic acid are dissolved in equal proportions in dimethylaminoid and the solution is added dropwise to water. The white cetrorelix pamoate precipitate with a peptide: pamoate kyas 2: 1 (mol / mol) ratio is filtered off and dried.
Example 3
Cetrorelix and pamoic acid in a molar ratio of 1: 1.6 are dissolved in a mixture of dimethylacetamide and optionally water, and the solution is added dropwise to water. The yellow precipitate is filtered off and dried. The precipitate obtained is made into a paste with 72% ethanol, dried at 35 ° C and sieved through a sieve with a mesh size of 80 to 125 pm.
Example 4
An alkaline embo-ala solution is added to the water-ethanolic solution of peptide acetate in a 2: 1 molar ratio of peptide embonate. The white precipitate is filtered off and dried. The dried solid is moistened with 50% ethanol, dried in a vacuum dryer and sieved. The white product contains peptide cEbooics with a ratio of 2: 1 (mol / mol).
Example 5
The alkaline embonikau solution is added to the water-ethanolic solution of peptide acetate in a molar ratio of peptide: embonic kyas 1: 1.6. The yellow precipitate is filtered off and. dried. The dried solid is moistened with 50% ethanol, dried in a vacuum dryer at 35 ° C and sieved. The yellow product contains peptide CEbooate with a ratio of 2: 1 (mol / mol) and excess embonic acid.

Experiments demonstrating the duration of action in animals:
Sediment suspensions were administered subcutaneously to male rats at a dose of 0.5 mg cetrorelix / kg body weight, and after administration the effect of the peptide on plasma testosterone was determined. The effect of cetrorelix is based on lowering testosterone levels. An injection suspension prepared according to DE 4023134 Al, which contained peptide pamoate in lactic and glycolic acid copolymers, was also tested as a reference. The period of action of cetrorelix in a form that has no prolonged effect was determined by testing the aqueous solution of cetrorelix acetate. Figure 3 is a graph of testosterone levels over 300 hours determined in male rats after administration of an aqueous solution of cetrorelix acetate (D-20761). The effect of testosterone suppression is obtained 6 hours after administration. Suppression to 1 mg / ml could still be seen in two animals for 24 hours and three animals for up to 48 hours or 2 days.
Figure 2 is a graph of testosterone levels for 300 hours in 4 animals (No. 11-14) after administration of the same dose of cetrorelix as the suspension of cetrorelix pamoate (D-20762) without thickening additives prepared according to example 1 (D-20762). Testosterone suppression was also obtained 6 hours after administration, with levels rising above 1 mg / ml in one animal after 192 hours (8 days), and the other three had suppression up to the 9th day.
Figure 4 is a graph of testosterone levels in rats treated with the embonate according to the invention (grain size: 80 pm -125 fm).
Comparison of figures 2 and 3 with figure 4 shows the advantages of the agent according to the invention.
Example 6
D-22213 acetate salt and pamoic acid are dissolved in dimethylacetamide in a 1: 3 molar ratio. The solution is added dropwise to water. The precipitate is filtered off and dried. The dry sludge is moistened with 70% ethanol, granulated, dried and sieved through a sieve with a sieve size from 80 to 125 pm. The resulting particles are filled into vials.
Example 7
The alkaline pamoate solution is added to an aqueous ethanolic solution of acetate D-22213. Peptide: pamoic acid molar ratio is 1: 3. Ethanol is removed in vacuo. An equivalent volume of isotonic aqueous mannitol solution is added to the ethanol-free suspension. The suspension is then freeze dried. The dried, frozen product can be resuspended in water as an injection suspension.
