Process for cross-linking hyaluronic acid to different polymers (other than hyaluronic acid)

Abstract

A process for the preparation of multiple cross-linked derivatives of hyaluronic acid (HA), comprising covalently cross-linking HA to one/more polymers other than HA via two/more different functional groups so that two/more chemically distinct cross-links are formed between HA and said polymer(s).

  1. 1
A process for the preparation of multiple cross-linked derivatives of hyaluronic acid, which process comprises covalently cross-linking HA to one or more polymers other than HA and optionally to another molecule of HA via two or more different functional groups, wherein said cross-linking is effected by contacting HA and a polymer other than HA with one or more chemical cross-linking agents so as to form two or more chemically distinct crosslinks between HA and said polymer.
  2. 5
A process according to any of claims 1 to 4 wherein the functional groups are selected from hydroxyl, carboxyl and amino.
  3. 6
A process according to any of claims 1 to 5 wherein the crosslinking is effected by means of two or more different bonds selected from ether, ester, sulfone, amine, imino and amide bonds.
  4. 7
A process according to any of claims 1 to 6 wherein the cross-linking agent is selected from formaldehyde, glutaraldehyde, divinyl sulfone, a polyanhydride, a polyaldehyde, a polyhydric alcohol, carbodiimide, epichlorohydrin, ethylene glycol, butanediol diglycidylether, diglycidylether, polyglycerol polyglycidylether, polyethylene glycol diglycidylether, polypropylene glycol diglycidylether, or a bis-or poly-epoxy cross-linker. Intellectual Property Office of NZ 1 7 JUL 2003
  5. 8
A process according to any of claims I to 7 wherein an ether bond is formed using a crosslinking agent selected from bis and poly epoxides under alkaline conditions. 5
  6. 9
A process according to any of claims 1 to 7 wherein an ester bond is formed using a crosslinking agent selected from bis and poly epoxides under acidic conditions.
  7. 11
A process according to any of claims 1 to 7 wherein an ether bond is formed using a glutaraldehyde cross-linking agent under acid conditions.
  8. 12
A process according to any of claims 1 to 11 wherein the crosslinking of each type of 15 functional group is effected sequentially.
  9. 15
A process according to any of claims 1 to 7 and 9 to 11 wherein the crosslinking of each type of functional group is effected simultaneously. 30
