Generally, pea protein hydrolysates for use in the nutritional products of the present disclosure are produced by: (1) blending a pea protein product that has at least 65% protein (by weight of dry matter) and water to form a mixture with a protein content of up to about 20%, preferably up to about 12%; (2) hydrolyze the mixture; and (3) filtering the hydrolyzed mixture to obtain the protein hydrolyzate.

Example 2
In this example, the hypoallergenic potential of the pea protein hydrolysates analyzed in Example 1 is evaluated and compared to the intact pea protein isolate (ISO) sample from Example 1 using hyperimmunization. of rabbits, in which rabbits are given the multiple immunizations for 35 days as will be described later. The immunogenicity of pea protein hydrolysates is then tested by ELISA (enzyme-linked immunosorbent assay), which measures the response of rabbit antigen-specific IgG to the intact pea protein / pea protein hydrolyzate immunogen.
The rabbits used in this example are put on a diet free of cow's milk, soy, and pea proteins for fourteen days prior to the first immunization. On day 0, the baseline antibody status of each rabbit was
IMPI evaluates with the blood sample of the corref to obtain the baseline, each rabbit is immunized with 5.0 mg of the equivalent amino acid content of pea protein included in 4.5 ml of antigen emulsified in complete Freund's adjuvant (CFA, for its acronym in English). Immunizations are given as follows: 20 x 0.1 ml per site / flank shaved intradermally, and 0.25 ml / popliteal area of the hind paw lymph node. On day 21, the second immunization is as follows: 0.50 ml / site X 2 sites / hind paw, intramuscular; and, 0.25 ml / site X 4 sites subscapularly. For each immunization, a constant dose of 5 mg amino acid equivalent of antigen is administered. The exsanguination of rabbits is on day 35 of the study.
A direct ELISA is used to quantify the antibody responses for each rabbit after hyperimmunization. Analysis optimization is achieved by using block titrations to determine antigen coating concentration and conjugate dilution. After optimization of assays, pea protein-specific rabbit antibodies are quantitated in serum samples from day 0 and day 35. Additionally, an immune response index (IRI units) is determined by dividing the normalized antibody titer on day 35 by the normalized baseline titer (day 0) on day 0 for each antiserum. The immunization groups contain 5-10 animals and the results for each animal are averaged. The

As shown in the table, pea protein hydrolysates show a wide range of antibody response. Low reactivity may indicate that pea protein hydrolysates still have epitopes that are recognized as antigens, but not immunogenic by rabbits. On the other hand, the pea protein hydrolysates BN, FB, NC, and CF showed significantly reduced immunogenicity after hyperimmunization relative to the other samples. The reduced immunogenicity of the pea protein hydrolyzate samples indicates that conserved pea antigens do not have the ability to stimulate an immune response.

In this analysis, the intact pea protein is coated on the ELISA plates. Incubation tubes containing the pea protein samples or solutions at standard concentrations plus anti-pea protein antibodies are prepared separately. After an incubation period, aliquots of the sample (or standard) plus the antibody solutions are added to the coated plates. After incubation and washing, an anti-rabbit IgG antibody conjugated to the enzyme is added. Plates are incubated, washed, and an enzyme substrate is added. A staining reaction occurs, which is inversely proportional to the concentration of pea antigen in the sample, which is quantitated according to a standard curve. INDUSTRIAL two area research sessions to characterize the taste of the pea protein hydrolyzate samples from Example 1 within the retort both in water and on an Alimentum® matrix (Abbott Laboratories, Columbus, Ohio). The Alimentum® matrix contains carbohydrates, fats, vitamins, and minerals to simulate the biochemistry of a hypoallergenic formula. It is used to determine the mutual organoleptic effects on the hydrolyzate and the finished formula relative to each other within the product matrix after retort treatment. The samples are compared with the current hydrolyzate (casein hydrolyzate) used in Alimentum® and the intact pea protein isolate (ISO) from example 1. In particular, it is known that hydrolyzed proteins are generally bitter, sour, salty, and / or fruity flavor. Therefore, it is difficult to incorporate this functional ingredient into nutritional formulations because it drastically damages the taste of the product.
