[Detailed Description of the Invention]
The present invention relates to the preparatively isolating method and aliquot apparatus which carry out ingredient separation of the blood by the sedimentation operation of gravity and which were improved. Blood consists of a plasma ingredient and blood cell ingredients, such as red corpuscles, white corpuscles, and blood platelets. Transfusion does not transfuse the blood which collected blood as it is, but divides it into these ingredient layers in recent years, and it transfuses more often only the specific ingredient which each patient needs, respectively. If blood is settled, a blood cell ingredient will precipitate gradually, and plasma will be separated, but the speed is slow. Therefore, it was not based on sedimentation by gravity, but the separation method using a centrifuge has been adopted widely conventionally. however, this method -- ^ -- a value centrifuge, that rotation power, a safeguard, etc. are needed. The result examined wholeheartedly that 1 uninvented persons should provide the device which can perform ingredient separation of blood efficiently by the sedimentation operation of gravity without using such artificial centrifugal force, Without using a centrifugal force place for blood level or by [ substantial ] passing continuously, giving the degree of climbing angle to the direction of a flow, blood was easily divided into the multilayer ingredient, scales were found out, and it completed as an invention of a separation method and a device, and has already applied. ('l? Wish 56-16190) These artificers, The result of having advanced examination wholeheartedly in order to obtain promptly Fractionation excluding red corpuscles from Yo Si and blood to the separation device of a smaller constituent of blood, adding [ at a surprising thing ]-into blood-bathing polymers V This -- very much -- effective -- hemagglutination -- promotion -- it finds out that n1 plasma ingredient is efficiently acquirable, and came to complete the present invention. That is, they are continuous to a tubular O channel, and one stratified or cIt Surely at least about the blood after adding water-soluble polymers into blood in a method of isolating an ingredient preparatively for blood by the sedimentation operation of gravity, in the present invention. Therefore, the space part which has the preparatively isolating method and blood feed port of a constituent of blood which consider as % mark making it shift in the style of [ which consists of an ingredient layer from which the blood style was separated ] a multilayer, and acquiring a request Q layer ingredient, In an aliquot apparatus of a constituent of blood using a constituent-of-blood separation machine which consists of at least one stratified or tubular blood channel which was open for free passage to the space part, and a space part which has an outlet of at least two constituents of blood which were open for free passage to the channel, Induction of blood, a mixer of the blood and solution containing water-soluble polymers, a constituent-of-blood preparative isolation machine, and a separated derivation part of a constituent of blood are the aliquot apparatus of a constituent of blood which makes it q# mold to have been arranged in this order. Not the patching method but the continuous point of the method of the present invention is the feature. As water-soluble polymers used by the present invention, water-soluble polyvinyl compounds, such as polysaccharide, such as gelatin or its derivative, dextran, a pull run, and inulin, or a derivative of those, polyvinyl alcohol, and polyvinyl pyrrolidone, or the derivative of those is used. As water-soluble polymers, a molecular weight is 1000. It is effective in Yo Si size coming and that of a potato accelerating condensation of red corpuscles. Since what has a very large molecular weight has antigenicity and does an obstacle to Re and the kidney, it is not preferred. and water-soluble larger polymers than 1000 smaller than molecular weight 100,000, more preferably water-soluble larger polymers smaller than molecule t50,000 than 1000 are used suitably. Water-soluble polymers have it, when the method of using as solution which has osmotic pressure almost equivalent to a physiological state performs ingredient separation of blood continuously. [ preferred ] As concentration of water-soluble polymers, 1~lO mass percent is desirable # 1-time range. As for the quantity of solution of the water-soluble polymers to add, it is preferred that it is 10~25 capacity percent to the introduced blood. the constituent-of-blood separation machine used for the present invention has a channel of blood inside -- [ in Si and its channel ] In order to be based on sedimentation rate, i.e., gravity, and also for the power of a from and the external force of the direction of a flow of blood to act on red corpuscles efficiently and to promote condensation lump formation of red corpuscles, a stratified channel -- the thickness -- below 20■ and Niji -- desirable -- 0.2~lO■, The cross-sectional area of Company is 3 (111 or less) at 0.5~5■ and a tubular channel preferably. As for the channel of the That separation machine with which the channel which is 0.001~0.5d still more preferably is used suitably 0.0003~1 m more preferably, it is preferred that the total volume has bigger capacity almost equivalent to the space part which has an outlet of a separation constituent of blood or than it. It is to distribute the space part which has a blood feed port to a channel as uniformly as possible, and for the blood introduced in the separation machine to flow through it, as long as it attains reed and this object, what kind of shape may be sufficient, but the thing small as much as possible of that capacity is desirable. As for the capacity of the separation machine, it is desirable that som is at least practical. When this device is used in order to perform extracorporeal circulation, it is desirable that it is below 500 WLt. In order to use the aliquot apparatus of the present invention suitably, it is desirable to shed the blood which added water-soluble polymers so that the holding time of the blood in the space part which has an outlet of the separation constituent of blood of the separation machine may become 1 minute or more. However, since the quantity of the separation ingredient which the blood volume which can be passed decreases and is obtained will fall if the holding time of the blood is too long not much, it is not desirable. As for a blood style, it is desirable to be warmed by 35~42degreeC, more preferably 37~40 degreeC. It is accelerated so that temperature is high, but condensation of red corpuscles is More! ll^There, hemolysis of red corpuscles, etc. take place and it is not desirable. The quality of the material of the device of the present invention must be the quality of the material which there are not 5 with required being a hard thing which does not change by internal pressure, and a poisonous elution thing, and cannot cause thrombus formation easily. The quality of the material of an airtight container needs to be a hard thing which does not change by internal pressure. It is required to be a nonpoisonous material of anti-Blood orange. However, an airtight container can be made from an elastic and thin material, and this can also be reinforced and used with hard material. Since it is easy to use this thing as a disposable container, it is a desirable embodiment, in view of a sanitary viewpoint. As a concrete container material, silicon resin, elastic vinylchloride resin, and polyethylene resin etc. are raised as polycarbonate resin, hard vinylchloride resin, an acrylic resin, aluminum, and an elastic thing. Next, a drawing explains the present invention concretely. Drawing 1 is an explanatory view showing the example of basic Fishing composition of the aliquot apparatus of the constituent of blood of the present invention. Drawing 2 is an example so that intelligibly [ in the mimetic diagram showing an example of a constituent-of-blood separation machine / 6 / the inside of 1. ]. If the present invention is explained according to the flow of blood now, the blood which was able to add the anticoagulant is introduced from blood induction 1, and if needed, with pump 2 like a roller pump, 3 carats of mixers will pass through it, and it will be conveyed to constituent-of-blood preparative isolation machine 4. With mixer 3, blood and the water-soluble polymers solution conveyed from storage machine 5 are mixed. Each constituent of blood separated with constituent-of-blood preparative isolation machine 4 is drawn from each derivation part 7 and 8. In the present invention, after blood is introduced into space part (A) 12 in the preparative isolation machine from blood feed port 11, constituent-of-blood preparative isolation machine 4 is sent succeeding one side of stratified blood channel 13, as the example of - is shown in Drawing 2. blood leaks and flows into a blood channel -- it being alike and following -- multi-blood platelet plasma -- food -- it separates into precipitation layer 15 which mainly turns into top Sengoku 16 from red 6 blood cell. Precipitation layer 15 may form the middle class in the state where a white corpuscle layer appears on on a red corpuscle layer, when separation further progresses. The ingredient separated in the lower stream of blood channel 13 lets outlet 17.18 which stagnated in space part (B) 14 and which carries out an opening into each ingredient layer pass, and is continuously discharged to derivation parts 7 and 8 using pump 9 if needed. A plurality of stratified channels as shown in Drawing 3 besides the above structures may be sufficient as a blood channel, and the singular number or a plurality of tubular channels may be sufficient as it. In order to control mixing of the ingredient of the precipitation layer to upper Placing discharged, it is desirable by using detection means lO (Drawing 1) by optical means etc. to adjust the discharge speed of each ingredient. The blood collecting machine usually according [ on the present invention and ] to a shunt, a hypodermic needle, etc. with blood induction, A pump is used together other lead pipes, a tap, and if needed, the useless device which introduces blood into a constituent-of-blood aliquot apparatus is said, and it is a constituent-of-blood derivation part, The device which derives each constituent of blood separated with the constituent-of-blood separation machine from a constituent-of-blood aliquot apparatus using a shaft, a tap, a blood-pressure control valve, intravenous drip, etc. a lead pipe and if needed is said. A mixer is for mixing the blood and water-soluble polymers solution which were able to add the anticoagulant agent, Although This which is by what can be completely mixed by churning etc. is desirable, even if it is a useless device which makes two fluid, such as a Y character base connector, join, the object can fully be achieved. Hereinafter, an example explains the mode of operation of the present invention in detail. Example 1 it is structure as shown in Drawing 2, using an acrylic resin as a constituent-of-blood separation machine -- thickness 2-1 of a blood channel -- capacity 15csi of the capacity 30-1 space part B. The separation machine which is total volume 50 Me of a separation machine was created. Created the constituent-of-blood aliquot apparatus as shown in Drawing 1 using the separation machine which kept it warm to 37 degreeC, solidification was made to prevent in Henocrine, the blood transfusion of the pig blood liquid warmed to 38 degreeC was carried out at the speed for 101 minutes, and the preparative isolation experiment was conducted. As water-soluble polymers, it is galley Husin (Toyo Jozo Co., Ltd. make; re-polymer 4111 of Gelatin decomposed materials, and sodium chloride 0.852 tlb) as a gelatin derivative. As salt Ichium calcium 0.0747 and polysaccharide, Dex F Rump 0 [dextran Sugariness (made in a large region factory) and 5 Thin dexterity/706% of grape II], dextran 40 [low-molecular dextran Sugariness (made in a large region factory), grape m1ss, and dextran 4010 Chi], and the 1st table book separation efficiency were defined as follows. It added. The result when the sedimentation rate in 37 degreeC used the pig blood liquid of 43- was shown in the 1st table. Since blood is not only separable into many ingredients, but a narrow sickroom can be easily enforced in the Sumiya power, without using L To and centrifugal force for using the device of the present invention as stated to details above, Sedimentation rate, such as systemic 1000 Tetratumus, is very useful to a plasma exchange therapy or a plasma purification treatment of a collagen disease patient, a cancer patient, etc. which rose comparatively slightly.
