    A process for the site selective genomic modification of yeasts of the species Pichia pastoris at a predetermined genomic site which comprises transforming a host strain of the species Pichia pastoris with a serially arranged linear DNA fragment comprising a first insertable DNA fragment, a construct comprising a selectable marker gene, a heterologous gene, wherein said heterologous gene, as well as said selectable marker gene, is positioned between the first insertable DNA fragment and a second insertable DNA fragment, and a regulatory region; wherein said heterologous gene is under the control of the regulatory region; and wherein the regulatory region-heterologous gene construct, in addition to said selectable marker gene, is positioned between the first insertable DNA fragment and a second insertable DNA fragment, a second insertable DNA fragment; wherein said first and second insertable DNA fragments are fragments having lengths in the range of about 200-5000 base pairs, of one of:the alcohol oxidase genes, or the histidinol dehydrogenase gene which are homologous with separate portions of the native Pichia genomic site at which genomic modification is to occur;wherein said first and second insertable DNA fragments are oriented with respect to one another in said linear DNA fragment as they are so oriented in the genome of Pichia pastoris;and wherein said construct is positioned between the first insertable DNA fragment and the second insertable DNA fragment.

    A process in accordance with claim 1, wherein said first and second insertable DNA fragments are characterized by the restriction map shown in FIGURE 1.

    A process in accordance with claim 1, wherein said first and second insertable DNA fragments are characterized by the restriction map shown in FIGURE 14.

    A serially arranged linear DNA fragment which comprises a first insertable DNA fragment, a construct comprising a selectable marker gene, and a heterologous gene, wherein said heterologous gene, as well as said selectable marker gene, is positioned between the first insertable DNA fragment and a second insertable DNA fragment and a regulatory region; wherein said heterologous gene is under the control of the regulatory region; and wherein the regulatory region-heterologous gene construct, in addition to said selectable marker gene, is positioned between the first insertable DNA fragment and the second insertable DNA fragment, and a second insertable DNA fragment; wherein said first and second insertable DNA fragments are fragments having lengths in the range of about 2005000־ base pairs, of one of:the alcohol oxidase genes, the histindinol dehydrogenase gene which are homologous with separate portions of the native Pichia genomic site at which genomic modification is to occur;wherein said first and second insertable DNA fragments are oriented with respect to one another in said linear DNA fragment as they are so oriented in the genome of Pichia pastoris;and wherein said construct is positioned between the first insertable DNA fragment and the second insertable DNA fragment.

    A linear DNA fragment in accordance with claim 7, r wherein said first and second insertable DNA fragments are characterized by the restriction map shown in FIGURE 1.

    A closed circular plasmid comprising a linear DNA fragment according to any one of claims 7 to 12 and bacterial plasmid DNA;wherein said bacterial plasmid DNA is positioned between the first insertable DNA fragment and the second insertable DNA fragment.
