    139 239746/4 WHAT IS CLAIMED IS:1. A method for generating a plant having improved resistance to an insect pest, the method comprising: (a) priming a seed prior to contacting the seed with an exogenous non-transcribable double-stranded RNA (dsRNA) molecule, wherein said priming is effected by: (i) washing the seed prior to said contacting;and (ii) drying the seed following step (i);(b) directly contacting the seed with an exogenous non-transcribable dsRNA molecule via soaking a seed of the plant in a solution comprising a concentration of 0.004 pg/pL to 1.5 pg/pL of the exogenous non-transcribable dsRNA molecule comprising a nucleic acid sequence that is identical or complementary to 18 or more contiguous nucleotides of an essential insect pest gene, or to the sequence of an RNA transcribed from the essential insect pest gene, wherein the seed is shaken in the solution for up to 24 hours and (c) germinating the seed to generate a plant exhibiting improved insect resistance after emerging from the seed, as compared to a control plant grown from a seed not contacted with the dsRNA molecule.

    The method of any one of Claims 1 or 2, wherein the essential insect pest gene is selected from the group consisting of V-ATPase, NADPH Cytochrome P450 Oxidoreductase, inhibitor of apoptosis (IAP), Chitin Synthase, EF1a, and β-actin.

    The method of any one of Claims 1-3, wherein the plant is resistant to Spodoptera littoralis, Diabrotica virgifera virgifera or Leptinotarsa decemlineata infestation. 140 239746/4

    A method of treating a seed to improve insect resistance of a plant grown from the seed, the method comprising:(a) priming a seed prior to contacting the seed with an exogenous non-transcribable double-stranded RNA (dsRNA) molecule, wherein said priming is effected by: (i) washing the seed prior to contacting;and (ii) drying the seed following step (i);and (b) introducing into the seed an exogenous non-transcribable dsRNA molecule comprising a sequence that is essentially identical or essentially complementary to at least 18 contiguous nucleotides of an essential insect pest gene or to the sequence of an RNA transcribed from the essential insect pest gene, via directly contacting the seed with the exogenous non-transcribable dsRNA molecule by soaking the seed in a solution comprising a concentration of 0.004 pg/pL to 1.5 pg/pL of the dsRNA molecule, wherein the seed is shaken in the solution for up to 24 hours, wherein the insect pest is selected from the group consisting of a Spodoptera spp, a Diabrotica spp. and a Leptinotarsa decemlineata, and wherein the plant grown from the seed exhibits improved insect resistance relative to a control plant.

    The method of any one of claims 1-19, wherein the exogenous nontranscribable dsRNA is present in the endosperm of the seed, the embryo of the seed, or both.

    The method of any one of claims 1-19, wherein the exogenous nontranscribable dsRNA is present at a higher concentration in the endosperm of the seed as compared to the embryo of the seed.

    The method of any one of claims 1-19, wherein the exogenous nontranscribable dsRNA is present at the same concentration in the embryo and endosperm of the seed.

    The method of any one of claims 1-23, wherein the exogenous nontranscribable dsRNA molecule comprises a nucleic acid sequence that is identical or complementary to at 18 or more contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs:21-26, 31, 34, 37, 38, 131-133, or 144-190.
