FIELD OF THE INVENTION

[0002] The present invention provides a salt form, and compositions thereof, useful as inhibitors of protein kinases.

BACKGROUND OF THE INVENTION

[0003] The search for new therapeutic agents has been greatly aided in recent years by a better understanding of the structure of enzymes and other biomolecules associated with diseases. One important class of enzymes that has been the subject of extensive study is protein kinases.

[0004] Protein kinases constitute a large family of structurally related enzymes that are responsible for the control of a variety of signal transduction processes within the cell. Protein kinases are thought to have evolved from a common ancestral gene due to the conservation of their structure and catalytic function. Almost all kinases contain a similar 250-300 amino acid catalytic domain. The kinases may be categorized into families by the substrates they phosphorylate (e.g., protein-tyrosine, protein-serine/threonine, lipids, etc.).

[0005] In general, protein kinases mediate intracellular signaling by effecting a phosphoryl transfer from a nucleoside triphosphate to a protein acceptor that is involved in a signaling pathway. These phosphorylation events act as molecular on/off switches that can modulate or regulate the target protein biological function. These phosphorylation events are ultimately triggered in response to a variety of extracellular and other stimuli. Examples of such stimuli include environmental and chemical stress signals (e.g., osmotic shock, heat shock, ultraviolet radiation, bacterial endotoxin, and H2O2), cytokines (e.g., interleukin-1 (IL- 1) and tumor necrosis factor a (TNF-a)), and growth factors (e.g., granulocyte macrophage- colony-stimulating factor (GM-CSF), and fibroblast growth factor (FGF)). An extracellular stimulus may affect one or more cellular responses related to cell growth, migration, 1 224171/2 differentiation, secretion of hormones, activation of transcription factors, muscle contraction, glucose metabolism, control of protein synthesis, and regulation of the cell cycle. US 2010/029610 relates to compounds of formulae I-a and I-b useful as inhibitors of protein kinases:

I-a I-b and also provides pharmaceutically acceptable compositions comprising said compounds and methods of using said compounds in the treatment of various disorders. US 7,718,662 describes kinase inhibitor compounds of formula

and methods for synthesizing such inhibitors and methods for using such inhibitors in the treatment of diseases. Further described are methods, assays and systems for determining an appropriate inhibitor of a protein, including a kinase.

Philip L. Gould (International Journal of Pharmaceutics; Vol. 33, p. 201-217, 1986) provides a rationale for salt selection for basic drugs using a Kepner-Tregoe decision analysis approach. The desired characteristics of the salt form, giving sufficient strength and toxicological suitability of the conjugate acid, are discussed on the basis of the various pivotal physicochemical properties, e.g., melting point, aqueous solubility and dissolution rate, stability and hydrophobicity. 2 224171/2

The Encyclopedia of pharmaceutical Technology (Salt Forms Pharmaceutical Aspect", Third edition OWEN I CORRIGAN, Informa Healthcare p. 3177-3187, 2006) summarizes the physicochemical aspects of salts (e.g. solubility, counterion pKa, etc.), the dissolution and biopharmaceutical aspects of salts and issues related to processing, formulation and selection of a salt form.

Byrn, S. et al. (Pharmaceutical Solids: A Strategic Approach to Regulatory Considerations; Pharmaceutical Research, Vol. 12, No. 7. P. 945-954, 1995) describes a conceptual approach to the characterization of pharmaceutical solids.

[0006] Many diseases are associated with abnormal cellular responses triggered by protein kinase-mediated events as described above. These diseases include, but are not limited to, autoimmune diseases, inflammatory diseases, bone diseases, metabolic diseases, neurological and neurodegenerative diseases, cancer, cardiovascular diseases, allergies and asthma, Alzheimer's disease, and hormone-related diseases. Accordingly, there remains a need to find protein kinase inhibitors useful as therapeutic agents.

SUMMARY OF THE INVENTION

[0007] It has now been found that the novel salt form of the present invention, and compositions thereof, is useful as an inhibitor of one or more protein kinases and exhibits desirable characteristics for the same. In general, this salt form, and pharmaceutically acceptable compositions thereof, is useful for treating or lessening the severity of a variety of diseases or disorders as described in detail herein.

DETAILED DESCRIPTION OF THE INVENTION

General Description of Certain Aspects of the Invention: [0018] United States published patent application number US 20100029610, published February 4, 2010 (“the ‘610 publication,”), describes certain 2,4-disubstituted pyrimidine compounds which covalently and irreversibly inhibit activity of one or more protein kinases, including Bruton's tyrosine kinase (“BTK”), a member of TEC-kinases. Such compounds include compound 1:

1 [0019] Compound 1 (N-(3-(5-fluoro-2-(4-(2-methoxyethoxy)phenylamino)pyrimidin-4-ylamino)phenyl)acrylamide) is designated as compound number I-182 and the synthesis of compound 1 is described in detail at Example 20 of the ‘610 publication.

[0020] Compound 1 is active in a variety of assays and therapeutic models demonstrating covalent, irreversible inhibition of BTK (in enzymatic and cellular assays). Notably, compound 1 was found to inhibit B-cell proliferation both in vitro and in vivo. Accordingly, compound 1 is useful for treating one or more disorders associated with activity of BTK.

[0021] It would be desirable to provide a salt form of compound 1 that, as compared to compound 1, imparts characteristics such as improved aqueous solubility, stability and ease of formulation. Accordingly, the present invention provides the besylate salt of compound 1. [0023] It will be appreciated by one of ordinary skill in the art that the benzensulfonic acid and compound 1 are ionically bonded to form compound 2. It is contemplated that compound 2 can exist in a variety of physical forms. For example, compound 2 can be in 3 224171/2 solution, suspension, or in solid form. In certain embodiments, compound 2 is in solid form.

When compound 2 is in solid form, said compound may be amorphous, crystalline, or a mixture thereof. Exemplary solid forms are described in more detail below.

[0024] In other embodiments, the present invention provides compound 2 substantially free of impurities. As used herein, the term “substantially free of impurities” means that the compound contains no significant amount of extraneous matter. Such extraneous matter may include excess benzensulfonic acid, excess compound 1, residual solvents, or any other impurities that may result from the preparation of, and/or isolation of, compound 2. In certain embodiments, at least about 95% by weight of compound 2 is present. In still other embodiments of the invention, at least about 99% by weight of compound 2 is present.

Solid Forms of Compound 2: [0027] It has been found that compound 2 can exist in a variety of solid forms. Such forms include polymorphs, solvates, hydrates, and amorphous. All such forms are contemplated by the present invention. In certain embodiments, the present invention provides compound 2 as a mixture of one or more solid forms selected from polymorphs, solvates, hydrates, and amorphous compound 2. 4 224171/2 [0028] As used herein, the term “polymorph” refers to the different crystal structures (of unsolvated forms) in which a compound can crystallize. As used herein, the term “solvate” refers to a crystal form with either a stoichiometric or non-stoichiometric amount of solvent is incorporated into the crystal structure. Similarly, the term “hydrate” refers to a crystal form with either a stoichiometric or non-stoichiometric amount of water is incorporated into the crystal structure.

[0029] In certain embodiments, compound 2 is a crystalline solid. In other embodiments, compound 2 is a crystalline solid substantially free of amorphous compound 2. Examples of suitable solvents useful in the present invention are a protic solvent, a polar aprotic solvent, or mixtures thereof. In certain embodiments, suitable solvents include an ether, an ester, an alcohol, a ketone, or a mixture thereof. In certain embodiments, the suitable solvent is methanol, ethanol, isopropanol, or acetone wherein said solvent is anhydrous or in combination with water or heptane. Uses, Formulation and Administration

Pharmaceutically Acceptable Compositions [0049] According to another embodiment, the invention provides a composition comprising Compound 2 and a pharmaceutically acceptable carrier, adjuvant, or vehicle. The amount of Compound 2 in compositions of this invention is such that is effective to measurably inhibit a protein kinase, particularly at least one of a TEC-kinase, or a mutant thereof, in a biological sample or in a patient. In certain embodiments, the amount of Compound 2 in compositions of this invention is such that is effective to measurably inhibit 9 224171/2 at least one of TEC-kinase, or a mutant thereof, in a biological sample or in a patient. In certain embodiments, a composition of this invention is formulated for administration to a patient in need of such composition. In some embodiments, a composition of this invention is formulated for oral administration to a patient.

[0052] Compositions of the present invention may be administered orally, parenterally, by inhalation spray, topically, rectally, nasally, buccally, vaginally or via an implanted reservoir. The term "parenteral" as used herein includes subcutaneous, intravenous, intramuscular, intra-articular, intra-synovial, intrasternal, intrathecal, intrahepatic, intralesional and intracranial injection or infusion techniques. Preferably, the compositions are administered orally, intraperitoneally or intravenously. Sterile injectable forms of the compositions of this invention may be aqueous or oleaginous suspension. The sterile injectable preparation may also be a sterile injectable solution or suspension in a non-toxic parenterally acceptable diluent or solvent, for example as a solution in 1,3-butanediol. Among the acceptable vehicles and solvents that may be employed are water, Ringer's solution and isotonic sodium chloride solution. In addition, sterile, fixed oils are conventionally employed as a solvent or suspending medium.

Suitable carriers include, but are not limited to, mineral oil, sorbitan monostearate, polysorbate 60, cetyl esters wax, cetearyl alcohol, 2-octyldodecanol, benzyl alcohol and water.

It will be appreciated that, although the general methods depict the synthesis of certain compounds of the present invention, the following general methods, and other methods known to one of ordinary skill in the art, can be applied to all compounds and subclasses and species of each of these compounds, as described herein.

General Procedures [00113] Powder X-ray diffraction patterns were obtained on a Bruker D8 Advance with

Cu-Ka radiation and LynxEye detector. The powder samples were deposited on a zero-background polished silicon sample holder and was rotated during measurement. Measurements performed as follows: 40 kV/40 mA tube power, 0.02° 2-theta step size, 37 second step time, and 2.5-50° 2-theta scanning range.

[00114] Proton Nuclear Magnetic Resonance (1H NMR) spectra were obtained on a Bruker model DPX-300 MHz NMR spectrometer. 1H NMR spectra were recorded at 300.13 MHz, using a 30 degree excitation pulse, with a pulse delay of 1 second, 16 scans. Deuterated DMSO was used as solvent.

Example 1

Preparation of Compound 2 (Form P1)

[00118] The besylate salt of Compound 1, i.e., Compound 2, was prepared as follows. Compound 1 was added to MTBE under nitrogen to form a slurry and the mixture heated to 50-55 °C. Benzenesulfonic acid, in MTBE, was added and the resulting mixture stirred for one hour. The mixture was cooled to 0-5 °C and allowed to stir for an hour. The resulting solids were collected by filtration and then dried at 65-70 °C under vacuum to afford Compound 2. Characterization of the resulting material demonstrated that the Compound 2 was crystalline and this crystalline form referred to as Form P1.

Example 3

Preparation of Compound 2 (Form P22) [00124] Compound 2 (82.2 mg) was suspended in methyl ethyl ketone (6 mL) and the suspension heated to 68 °C while adding 8 mL methyl ethyl ketone. A clear solution was obtained and heated to 75 °C. The solution was cooled to 5 °C and solvent partially evaporated to obtain a white precipitate. The resulting solid was recovered by filter centrifugation to afford form P22. The material was characterized and the results as follows: [00125] The PXRD pattern of Compound 2 Form P22, as compared to Form P1, is depicted in Figure 6. The FT-Raman spectrum of Form P22 is depicted in Figure 7.. The TG-FTIR spectrum of Form P22 is depicted in Figure 8. The 1H NMR of Form P22 is 27 224171/2 depicted in Figure 9 and is consistent with the structure of Compound 2 having 1:1 ratio with its besylate salt. The DSC thermogram is depicted in Figure 10 and shows a single endothermic event at 193.7 °C.
