31 claims: 15 independent, 16 dependent

  1. 1
Claims:1. A chlorotoxin conjugate comprising: (i) a chlorotoxin peptide comprising an amino acid sequence set forth in SEQ ID NO: 2, in which Lysl5 and Lys23 are substituted by an amino acid other than lysine to provide a modified chlorotoxin having a single lysine residue, Lys27;and (ii) a fluorescent label selected from DyLight-680, DyLight-750, VivoTag-750, DyLight800, VivoTag-680, and indocyanine green;wherein the chlorotoxin peptide is covalently coupled to the fluorescent label through the lysine residue, and said chlorotoxin peptide binds to chlorotoxin binding sites expressed by cancer cells.
  2. 2
A chlorotoxin conjugate comprising a chlorotoxin peptide covalently coupled to a fluorescent label selected from DyLight-680, DyLight-750, VivoTag-750, DyLight-800, VivoTag-680, and indocyanine green, wherein the chlorotoxin peptide comprises an amino acid sequence corresponding to that of native chlorotoxin (SEQ ID NO:1) in which Lysl5, Lys23, or a combination thereof are substituted with an amino acid other than lysine.
  3. 8
A composition comprising a chlorotoxin conjugate of any one of Claims 1 to 7.
  4. 10
A pharmaceutical composition comprising a chlorotoxin conjugate of any one of Claims 1 to 7 for use in treating a disease or condition treatable by administering chlorotoxin, in a subject in need thereof.
  5. 11
The chlorotoxin conjugate of any one of Claims 1 to 7, wherein the chlorotoxin peptide is covalently coupled to one or more of a therapeutic, diagnostic, imaging, or targeting agent, or a moiety that increases the circulatory half-life of the chlorotoxin conjugate. 222930/3
  6. 14
The chlorotoxin conjugate of any one of Claims 11 to 13, wherein the diagnostic or imaging agent is selected from the group consisting of a quantum dot or polymeric dot.
  7. 15
The chlorotoxin conjugate of any one of Claims 11 to 13, wherein the diagnostic or imaging agent is selected from the group consisting of a boron nanoparticle, a boron and carbon nanoparticle, a boron carbide nanoparticle, boron-containing polymer, a boron and carbon containing polymer, a boron carbide polymer, and any of these nanoparticles or polymers further comprising gadolinium.
  8. 21
A method for imaging a tissue imagable by chlorotoxin, comprising contacting a tissue with the chlorotoxin conjugate of any one of Claims 1-16 to image a tissue imagable by chlorotoxin.
  9. 22
A method for detecting cancer detectable by chlorotoxin, comprising contacting a tissue with the chlorotoxin conjugate of any one of Claims 1-16 to detect cancer detectable by chlorotoxin.
  10. 23
A chlorotoxin conjugate of any one of Claims 1-16 for use in a method for detecting and removing cancer detectable by chlorotoxin, wherein said method comprises:(a) contacting a tissue with said chlorotoxin conjugate to detect cancerous tissue, and (b) removing the cancerous tissue detected by the chlorotoxin conjugate. 222930/3
  11. 24
A pharmaceutical composition comprising a chlorotoxin conjugate of any one of Claims 1 -20 for use in a method for treating cancer targeted by a chlorotoxin conjugate, wherein said method comprises contacting a tissue that binds to modified chlorotoxin with said composition.
  12. 25
The chlorotoxin conjugate of any one of Claims 1-16 and 20, for use in:(a) detecting cancer cells in vivo', (b) detecting and removing cancer cells in vivo', (c) detecting a cancerous or tumor tissue in an individual, wherein the chlorotoxin conjugate is for administration to the individual such that the chlorotoxin conjugate binds to the cancerous or tumor tissue;or (d) imaging cancerous or tumor tissue m an individual, wherein the chlorotoxin conjugate is for administration to the individual such that the chlorotoxin conjugate binds to the cancerous or tumor tissue.
  13. 26
Use of the chlorotoxin conjugate of any one of Claims 1-16 and 20 for the manufacture of a medicament for:(a) detecting or imaging cancer cells in vivo;or (b) detecting and removing, or imaging and removing cancer cells in vivo.
  14. 28
An in vitro or ex vivo method for detecting or imaging cancerous tissue with a chlorotoxin conjugate or a composition thereof, wherein the chlorotoxin conjugate comprises the chlorotoxin conjugate of any one of Claims 1-16 and 20, said method comprising:(a) contacting a tissue with the chlorotoxin conjugate or the composition thereof;and (b) analyzing the tissue to detect the binding of the chlorotoxin conjugate to cancer cells within the tissue.
