  1. 1
216873/5 57 Claims 1. A method for isolating a recombinant protein that is insoluble in the cytoplasm of a host cell and is not found as an inclusion body, comprising, a) separating an insoluble recombinant protein not found as an inclusion body from soluble host cell proteins;and b) solubilising the separated recombinant protein, wherein the recombinant protein is a C383S or C383A mutant of an oxalate decarboxylase (OxDC) wild type recombinant protein according to SEQ ID NO. 1 or SEQ ID NO. 2, or a protein encoded by a sequence selected from SEQ. ID NO. 3, SEQ. ID NO. 4, SEQ. ID NO. 5, SEQ. ID NO. 6, SEQ. ID NO. 7, SEQ. ID NO. 8, SEQ. ID NO. 15, SEQ. ID NO. 16, SEQ. ID NO. 17, SEQ. ID NO. 18 or SEQ. ID NO. 19.
  2. 4
A method according to any one of the preceding claims, wherein the solubilising of the recombinant protein comprises adding binding ligands or providing a pH in which the protein is soluble.
  3. 5
A method according to any one of claims 2-4, wherein the isolating comprises precipitating the recombinant protein out of the solubilizing solution.
  4. 7
A method according to any one of claims 1-6, wherein the recombinant protein is the C383S mutant of the OxDC wild type recombinant protein or a protein encoded by a sequence selected from SEQ. ID NO. 3, SEQ. ID NO. 4, SEQ. ID NO. 5, SEQ. ID NO. 6, SEQ. ID NO. 7 or SEQ. ID NO. 8.
  5. 8
A method according to any one of claims 1-6, wherein the recombinant protein is the C383A mutant of the OxDC wild type recombinant protein or a protein encoded by a sequence selected from SEQ. ID NO. 15, SEQ. ID NO. 16, SEQ. ID NO. 17, SEQ. ID NO. 18 or SEQ. ID NO. 19. 216873/5 58
  6. 9
Spray-dried particles comprising one or more recombinant proteins, selected from the group consisting of the C383S, and the C383A mutant of the oxalate decarboxylase (OxDC) wild type recombinant protein according to SEQ. ID NO. 1 or SEQ. ID NO. 2, and a protein encoded by a sequence selected from SEQ. ID NO. 3, SEQ. ID NO. 4, SEQ. ID NO. 5, SEQ. ID NO. 6, SEQ. ID NO. 7, SEQ. ID NO. 8, SEQ. ID NO. 15, SEQ. ID NO. 16, SEQ. ID NO. 17, SEQ. ID NO. 18, or SEQ. ID NO. 19, and a polymeric material.
  7. 12
Spray-dried particles according to any one of claims 9-11, wherein the activity of the one or more recombinant protein at the most decreases to 30% when incubated in a 3.2 mg/ml pepsin solution having a pH of 3.2 for 40 minutes with the initial activity being set to 100%.
  8. 13
Spray-dried particles according to any one of claims 9-12, wherein the polymeric material is a poly(meth)acrylate.
  9. 14
A composition comprising spray-dried particles as defined in any one of claims 9 13.
  10. 17
Use of a composition comprising spray-dried particles comprising one or more oxalate reducing enzymes and a polymeric material according to any one of claims14- 216873/5 59 16, in the manufacture of a medicament for reducing absorption of oxalate present in the stomach, and/or reducing at least a portion of the oxalate.
  11. 20
A composition according to any one of claims 14-16 for use in reducing absorption of oxalate present in the stomach.
  12. 21
Use of a recombinant protein encoded by any DNA sequence selected from SEQ. 20 ID NO. 3, SEQ. ID NO. 4, SEQ. ID NO. 5, SEQ. ID NO. 6, SEQ. ID NO. 7, SEQ. ID NO. 8, SEQ. ID NO. 15, SEQ. ID NO. 16, SEQ. ID NO. 17, SEQ. ID NO. 18 and SEQ. ID NO. 19 for the manufacture of a medicament for reducing absorption of oxalate present in the stomach.
