A method of preparing a compound having general Formula II: Formula II wherein: A, is CH2;A2 is CH2;R, is (CH2)14CH3;R2 is whereas: X is CH2;Y is hydrogen;and OH O- Z is \ ;and R3 is selected from the group consisting of hydrogen and phosphoryl choline, the method comprising: 77 199792/2 reacting a first compound comprising a glycerolic backbone and a free hydroxyl group, said first compound having general Formula I: Formula I;wherein: A, is CH2;Ri is (CH2)14CH3;and R-3 is a protecting group;and a second compound, said second compound being a 1-hexene having a leaving group at position 6 thereof for forming an ether bond with said free hydroxyl group of said first compound, to thereby obtain a third compound, said third compound having said glycerolic backbone and an unsaturated bond-containing residue being attached to said glycerolic backbone via the ether bond;collecting and purifying said third compound, thereby obtaining a purified third compound;reacting said purified third compound with an oxidizing agent in a one-step procedure, so as to oxidize said unsaturated moiety to form said Z, and removing said protecting group, to thereby obtain a compound of Formula II, which is 1-hexadecyl-2-(4'-carboxy)butyl-glycerol having formula: O(CH2)15CH3 0. OH OH or a salt thereof;and 78 199792/2 isolating said l-hexadecyl-2-(4'-carboxy)butyl-glycerol to thereby obtain purified 1 -hexadecyl-2-(4'-carboxy)butyl-glycerol.

    The method of any one of claims 1-2, wherein said purifying of said third compound comprises:contacting said third compound with a solvent to form a mixture, wherein said third compound is soluble in said solvent, whereby impurities formed during said reacting of said first compound and said second compound are insoluble in said solvent, wherein said mixture comprises said third compound in said solvent and insoluble impurities;removing said insoluble impurities;and removing said solvent.

    The method of any one of claims 1-4, wherein said oxidizing agent comprises a mixture of a periodate and a permanganate.

    The method of any one of claims 1-6, wherein said reacting said purified third compound with an oxidizing agent is effected in the presence of a base.

    The method of any one of claims 1-8, wherein removing said protecting group and isolating said l-hexadecyl-2-(4’-carboxy)butyl-glycerol are performed within the same synthetic step. 79 199792/2

    The method of any one of claims 1-10, wherein said protecting group is trityl.

    The method of any one of claims 1-11, further comprising:reacting said purified l-hexadecyl-2-(4’-carboxy)butyl-glycerol with a reactive phosphorus-containing compound subsequent to converting said Z of said 1 -hexadecyl-2-(4’-carboxy)butyl-glycerol to an ester moiety, to thereby provide a compound having a reactive phosphorus-containing group attached to the glycerolic backbone;and converting said reactive phosphorus-containing group to said phosphoryl choline;and hydrolyzing said ester moiety, thereby obtaining a compound of general Formula II, which is l-hexadecyl-2-(4’-carboxy)butyl-glycero-3-phosphocholine having the formula: CH3 OH 80 199792/2 or a salt thereof. 1'3. The method of claim 12, wherein said reactive phosphorus-containing compound is phosphorus oxychloride (POCI3).

    The method of any one of claims 12-13, wherein converting said reactive phosphorus-containing group to said phosphoryl choline comprises reacting said reactive phosphorus-containing group with ethanolamine and methyltosylate.

    The method of any one of claims 12-14, wherein said converting said Z to an ester moiety comprises reacting said l-hexadecyl-2-(4’-carboxy)butyl-glycerol with methanol to form a methyl ester.
