Method for evaluating an inflammatory condition using calibrated gene expression profiles

Abstract

The invention provides a method, for evaluating a biological condition of a subject, comprising: a. obtaining from the subject a sample having at least one of RNAs and proteins; b. deriving from the sample a first profile data set, the first profile data set including a plurality of members, each member being a quantitative measure of the amount of a distinct RNA or protein constituent in a panel of constituents selected so that measurement of the constituents enables measurement of the biological condition; and c. producing a calibrated profile data set for the panel, wherein each member of the calibrated profile data set is a function of a corresponding member of the first profile data set and a corresponding member of a baseline profile data set for the panel, the calibrated profile data set providing a measure of the biological condition of the subject.

    A method for evaluating an inflammatory condition of a subject, based on a first sample obtained from the subject, the sample providing a source of RNAs; characterised in that the method comprises:(a) deriving from the first sample a first profile data set, which first profile data set comprises 3 or more members, each member being a relative quantitative measure of the amount in said first sample of a distinct transcribed RNA constituent in a panel of constikients selected such that measurement of said constituents enables evaluation of said inflammatory condition, the relative quantitation of mRNA in said sample being determined by performing quantitative PCR on RNA extracted from the sample with a defined efficiency of amplification for the target transcripts and determining the difference in threshold cycles (delta Ct) between a size marker and the target transcripts of interest;and (b) producing a calibrated profile data set for the panel, each member of the calibrated profile data set being a function of a corresponding member of the first profile data set and a corresponding member of a baseline profile data set for the panel;wherein the calibrated profile data set represents a set of values in which a pattern of variation correlates in a reproducible manner with a particular condition.

    A method according to any one of claims 1-4, wherein the condition is characterised by inflammation and the panel includes at least one of IL-1 alpha, IL-1 beta, IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, IL2p40, IL-18, GMCSF, IFN-gamma, TGF-b, cox-1, cox-2, ICE, mmp-3, mmp-9, ICAM, TNF-a and TNF-b.

    A method according to any of claims 1 to 4, wherein the condition is characterized by liver metabolism and the panel of constituents is a liver metabolism panel.
