WE CLAIM :1. A recombinant nucleic acid construct comprising: (i) nucleic acid encoding lymphotoxin as depicted in Fig. 2A hereof, or (ii) nucleic acid as obtainable by hybridization with that depicted in Fig. 2A hereof, and encoding a, lymphotoxin polypeptide: or (iii) nucleic acid encoding a lymphotoxin polypeptide which has substanlial structural homology with at least a portion of the lymphotoxin amino acid sequence depicted in Fig. 2A hereof;said lymphotoxin polypeptide in (ii) or (iii) displaying preferential cytotoxic activity of, and/or immunological cross-reactivity with, and/or the ability to compete for lymphotoxin cell surface receptors with, cytotoxic lymphotoxin.
  2. 4
A recombinant nucleic acid construct according to any one of claims 1. 2 and 3 wherein the lymphotoxin amino acid sequence encoded differs from that of Fig. 2A by virtue of amino acid deletion, substitution or insertion.
  3. 5
A recombinant nucleic acid construct according to any one of claims 14־ wherein said nucleic acid encodes leucyl amino-terminal lymphotoxin.
  4. 6
A recombinant nucleic acid construct according to any one of claims 15־ wherein said nucleic acid encodes a human allele of the Fig. 2A sequence.
  5. 7
A recombinant nucleic acid construct according to any one of claims 15־ wherein the nucleic acid encodes a non-human animal lymphotoxin.
  6. 12
A recombinant nucleic acid construct according to claim It wherein said site comprises lys־lys or lysarg.
  7. 13
A recombinant nucleic acid construct according to any one ol claims 10 to 12, wherein the encoded fusion product is cytolytically inactive prior to proteolytic hydrolysis.
  8. 32
A recombinant nucleic acid construct according to any one of the preceding claims, in the lorm of a replicable vector.
  9. 40
A method according to daim 39 wherein the cell is a yeast or a non-human mammalian cell, said nucleic acid encodes human prelymphotoxin, and lymphotoxin having variant glycosylation with respect 10 to lymphotoxin produced in homologous cellsis recovered from the culture.
  10. 41
A method according to daim 39 wherein the cell is a prokaryote.
  11. 42
A polypeptide as obtainable by the method of any one of claims 39 to 41, which is unglycosylated or is has variant glycosylation with respect to lymphotoxin produced in homologous cells, and which displays preferential cytotoxic activity of cytotoxic lymphotoxin and/or the ability to compete with cytotoxic lymphotoxin for lymphotoxin cell surface receptors.
  12. 43
A polypeptide as obtainable by the method of any one of claims 39 to 41 which is unglycosyiated or 20 has variant glycosylation and which has an amino add sequence differing from that depicted in Fig. 2A hereof by virtue of amino add deletion, substitution or insertion.
  13. 45
A process which comprises, following the method of any one of claims 39 to 41, the use of the polypeptide expression product in the preparation of a medicament.
  14. 46
A diagnostic or screening probe (or lymphotoxin nucleic acid comprising nucleic add which is hybridizable with the nucleic add depicted in Fig. 2A hereof, and covalently labelled with a detectable 30 substance. 4
  15. 47
A polypeptide as obtainable by the method of any one of claims 39-41 which is unglycosylated or has variant glycosylation in glutaraldehyde-po|ymerised with respect to lymphotoxin produced in homologous cells form or in the form of an adsorption complex with alum.
