Methods of preparation and purification of a compound, intermediates thereof, a polymorph thereof, and related compounds are disclosed. Formulations and uses thereof in the treatment of LFA-1 mediated diseases are also disclosed.

    A method of synthesizing a compound of Formula I:or a salt thereof, comprising the steps: a) performing hydrolysis of Formula AA with a base under biphasic conditions: wherein R is a carbon-containing moiety or a silyl-containing moiety;and b) isolating said compound of Formula I or a salt thereof.

    A reaction mixture comprising a compound of Formula I:or a salt thereof, and further comprising a compound of Formula AA wherein R is a carbon-containing moiety or a silyl-containing moiety.

    A method of purifying a compound of Formula I:or a salt thereof, comprising the steps: a) obtaining crude compound of Formula I or a salt thereof and recrystallizing said crude compound with aqueous acetone;and b) isolating said compound of Formula I or a salt thereof by removal of aqueous acetone.

    A solid composition comprising a pharmaceutically acceptable carrier and further comprising the compound of Formula I:wherein said compound is polymorph Form II wherein the polymorph is characterized as having an X-ray powder diffraction pattern with peaks at a reflection angle 20 of about 10.8, 16.4, and 21.8 degrees.
