Therapeutic and diagnostic methods for cancer

Abstract

The present invention provides therapeutic and diagnostic methods and compositions for cancer, for example, bladder cancer. The invention provides methods of treating bladder cancer, methods of determining whether a patient suffering from bladder cancer is likely to respond to treatment comprising a PD-L1 axis binding antagonist, methods of predicting responsiveness of a patient suffering from bladder cancer to treatment comprising a PD-L1 axis binding antagonist, and methods of selecting a therapy for a patient suffering from bladder cancer, based on expression levels of a biomarker of the invention (e.g., PD-L1 expression levels in tumor-infiltrating immune cells in a tumor sample obtained from the patient) and/or based on the determination of a tumor sample subtype.

    An effective amount of a PD-L1 binding antagonist for use in treating a patient suffering from a locally advanced or metastatic urothelial carcinoma who has had disease progression during or after platinum-based chemotherapy, wherein the PD-L1 binding antagonist is atezolizumab.

    The PD-L1 binding antagonist for use of any one of claims 1-5, wherein a tumor sample obtained from the patient has been determined to have a detectable level of PD-L1 in tumor-infiltrating immune cells that comprise about 1% or more of the tumor sample, and wherein the level of PD-L1 is a protein expression level.

    The PD-L1 binding antagonist for use of any one of claims 1 to 8, wherein the tumor sample is a formalin-fixed and paraffin-embedded (FFPE) tumor sample.
