15 claims: 3 independent, 12 dependent

  1. 1
A method of amplifying a target polynucleotide in a sample, the method comprising:combining a sample and nucleic acid polymer particles in an amplification reaction mixture, each such particle comprising a non-nucleosidic polymer network having attached throughout its volume a primer at a concentration of at least 1 × 10 5 primers per (µm 3 , a region of the target polynucleotide capable of annealing to the primer, the volumes of the polymer networks have a coefficient of variation of fifteen percent or less, the non-nucleosidic polymer network including a polyacrylamide gel having a total monomer percentage in a range of 3% to 20% and being permeable to proteins having a size in the range of 50 kilodaltons to 200 kilodaltons;and performing an amplification reaction so that primers on a nucleic acid polymer particle are repeatedly extended along a target polynucleotide annealed thereto so that a clonal population of complements of such target polynucleotide is formed on the polymer networks.
  2. 14
The method of one or more of claims 1 to 13, wherein the particle comprising a non-nucleosidic polymer network has attached throughout its volume a second primer at a concentration of at least 1 × 10 5 primers per µm 3 , a region of the target polynucleotide capable of annealing to the primer.
  3. 15
The method of one or more of claims 1 to 14, further comprising:disposing the nucleic acid polymer particles on a planar array;and performing a sequencing reaction simultaneously on the clonal populations on the nucleic acid polymer particles.
