15 claims: 5 independent, 10 dependent

  1. 1
A nucleotide analog comprising:a) a nucleotide comprising a phosphate moiety, a base, and a sugar;b) an electrochemically detectable, photocleavable terminating moiety attached to the nucleotide;and c) a second photocleavable terminating moiety attached to the nucleotide.
  2. 3
The nucleotide analog of any one of claims 1 to 2 wherein the electrochemically detectable, photocleavable terminating moiety is attached to the nucleotide by a photocleavable linker.
  3. 10
A composition comprising a nucleotide analog of any one of claims 1 to 9 and a polymerase.
  4. 11
Use of a nucleotide analog of any one of claims 1 to 9 in a nucleic acid sequencing reaction.
  5. 12
A method for sequencing a nucleic acid, the method comprising:a) hybridizing a primer to a nucleic acid to form a hybridized primer/nucleic acid complex;b) providing a plurality of nucleotide analogs, each nucleotide analog comprising a nucleotide, an electrochemically detectable, photocleavable terminating moiety attached to the nucleotide and a second photocleavable terminating moiety attached to the nucleotide;c) reacting the hybridized primer/nucleic acid complex and the nucleotide analog with a polymerase to add the nucleotide analog to the primer by a polymerase reaction to form an extended product comprising an incorporated nucleotide analog;d) querying the extended product to identify the incorporated nucleotide analog;e) exposing the extended product to a light source providing a wavelength of light to remove an electrochemically detectable, photocleavable terminating moiety from the incorporated nucleotide analog;and f) exposing the extended product to a light source providing a second wavelength of light to remove a second photocleavable terminating moiety from the incorporated nucleotide analog.
