15 claims: 9 independent, 6 dependent

  1. 1
A nanoparticle complex comprising:a nanoparticle core;a number of antigen-MHC complexes (pMHC) that comprise a number of MHC proteins and a number of multiple sclerosis-related antigens;and wherein the nanoparticle complex has an antigen-MHC density from 0.05 molecules pMHC/100 nm 2 to 30 molecules pMHC/100 nm 2 , and wherein the nanoparticle core optionally comprises a metal, a metal oxide, a metal sulfide, a metal selenide, a magnetic material, a polymer, iron, iron oxide, or gold, and wherein the nanoparticle core is optionally coated with dextran, mannitol, and polyethylene glycol.
  2. 4
The nanoparticle complex of claims 1 to 3, wherein the nanoparticle is non-liposomal.
  3. 5
The nanoparticle complex of claims 1 to 3, wherein the antigen-MHC complex is non-covalently linked to the nanoparticle core or the antigen-MHC complex is covalently linked to the nanoparticle core optionally through a linker less than 5 kD in size, and wherein the linker optionally comprises polyethylene glycol.
  4. 6
The nanoparticle complex of claims 1 to 3, wherein the MHC of the antigen-MHC-nanoparticle complex is a MHC class I or II, wherein MHC class I optionally comprises all or part of a HLA-A, HLA-B, HLA-C, HLA-E, HLA-F, HLA-G or CD-1 protein and MHC class II optionally comprises all or part of a HLA-DR, HLA-DQ, or HLA-DP protein.
  5. 10
A composition comprising a carrier and a therapeutically effective amount of the nanoparticle complex of any one of claims 1 to 9
  6. 11
A method for making, preparing or obtaining the nanoparticle complex of any one of claims 1-9, comprising coating or complexing a number of multiple-sclerosis related antigen-MHC complexes onto a nanoparticle core, wherein:the core optionally comprises a metal, a metal oxide, a metal sulfide, a metal selenide, a magnetic material, a polymer, iron, iron oxide, or gold;wherein the nanoparticle core is optionally coated with dextran, mannitol, and polyethylene glycol;and wherein the antigen-MHC complexes (pMHC) are coated onto the nanoparticle core at a density from 0.05 molecules pMHC/100 nm 2 to 30 molecules pMHC/100 nm 2 .
  7. 12
The nanoparticle complex of any one of claims 1-9, for use in expanding and/or developing populations of anti-pathogenic autoreactive T-cells in a subject with multiple sclerosis or a multiple sclerosis-related disorder.
  8. 13
The nanoparticle complex of any one of claims 1-9, for use in treating multiple sclerosis or a multiple sclerosis related disorder in a subject in need thereof.
  9. 15
A kit comprising the complex of any one of claims 1-9, and instructions for use.
