Antigen binding proteins to proprotein convertase subtilisin kexin type 9 (PCSK9)

Abstract

Antigen binding proteins that interact with Proprotein Convertase Subtilisin Kexin Type 9 (PCSK9) are described. Methods of treating hypercholesterolemia and other disorders by administering a pharmaceutically effective amount of an antigen binding protein to PCSK9 are described. Methods of detecting the amount of PCSK9 in a sample using an antigen binding protein to PCSK9 are described.

    An antigen binding protein which (i) comprises (a) a light chain variable domain comprising an amino acid sequence at least 80% identical to an amino acid sequence selected from the group consisting of SEQ ID NOs 481, 477, and 473, and(b) a heavy chain variable domain comprising an amino acid sequence at least 80% identical to an amino acid sequence selected from the group consisting of SEQ ID NOs:479, 475, and 471;(ii) comprises the CDRL1, CDRL2, and CDRL3 sequences of an amino acid sequence selected from the group consisting of SEQ ID NOs 481, 477, and 473, and the CDRH1, CDRH2, and CDRH3 sequences of an amino acid sequence selected from the group consisting of SEQ ID NOs 479, 475, and 471;or(iii) comprises a CDRL1, CDRL2, and CDRL3 sequence encoded by a nucleic acid that hybridizes to the nucleic acid encoding the CDRL1, CDRL2, and CDRL3 sequences of SEQ ID NOs: 481, 477 or 473, respectively;and a CDRH1, CDRH2, and CDRH3 sequence encoded by a nucleic acid that hybridizes to the nucleic acid encoding the CDRH1, CDRH2, and CDRH3 sequences of SEQ ID NOs: 479, 475, or 471, respectively, the hybridization optionally being carried out under moderately stringent conditions, wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of the preceding claims, comprising:(a) a light chain variable domain comprising an amino acid sequence at least 95% identical to an amino acid sequence selected from the group consisting of SEQ ID NOs 481, 477, and 473, and(b) a heavy chain variable domain comprising an amino acid sequence at least 95% identical to an amino acid sequence selected from the group consisting of SEQ ID NOs: 479, 475, and 471;wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of the preceding claims, comprising:(a) a light chain variable domain comprising an amino acid sequence at least 97% identical to an amino acid sequence selected from the group consisting of SEQ ID NOs 481, 477, and 473, and(b) a heavy chain variable domain comprising an amino acid sequence at least 97% identical to an amino acid sequence selected from the group consisting of SEQ ID NOs: 479, 475, and 471;wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of the preceding claims, comprising:(a) a light chain variable domain comprising an amino acid sequence selected from the group consisting of SEQ ID NOs 481, 477, and 473, and(b) a heavy chain variable domain comprising an amino acid sequence selected from the group consisting of SEQ ID NOs: 479, 475, and 471;wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of the preceding claims, wherein the light chain variable domain and the heavy chain variable domain are selected from the group of combinations consisting of:a light chain variable domain having the sequence set forth in SEQ ID NO: 481 and a heavy chain variable domain having the sequence set forth in SEQ ID NO: 479;a light chain variable domain having the sequence set forth in SEQ ID NO: 477, and a heavy chain variable domain having the sequence set forth in SEQ ID NO: 475;and a light chain variable domain having the sequence set forth in SEQ ID NO: 473 and a heavy chain variable domain having the sequence set forth in SEQ ID NO:471, wherein the antigen binding protein specifically binds to the PCSK9.

    The antigen binding protein of any of preceding claims 1, 2 or 3, comprising (a) a light chain variable domain comprising an amino acid sequence at least 90% identical to the amino acid sequence of SEQ ID NOs 481,(b) a heavy chain variable domain comprising an amino acid sequence at least 90% identical to the amino acid sequence of SEQ ID NOs:479, wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of preceding claims 1, 2, 3 or 8, comprising (a) a light chain variable domain comprising an amino acid sequence at least 95% identical to the amino acid sequence of SEQ ID NOs 481,(b) a heavy chain variable domain comprising an amino acid sequence at least 95% identical to the amino acid sequence of SEQ ID NOs:479, wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of preceding claims 1, 2 or 3, comprising (a) a light chain variable domain comprising an amino acid sequence at least 90% identical to the amino acid sequence of SEQ ID NOs 477,(b) a heavy chain variable domain comprising an amino acid sequence at least 90% identical to the amino acid sequence of SEQ ID NOs:475, wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of preceding claims 1, 2 or 3, comprising (a) a light chain variable domain comprising an amino acid sequence at least 90% identical to the amino acid sequence of SEQ ID NOs 473,(b) a heavy chain variable domain comprising an amino acid sequence at least 90% identical to the amino acid sequence of SEQ ID NOs:471, wherein the antigen binding protein specifically binds to PCSK9.

    The antigen binding protein of any of the preceding claims, wherein the antigen binding protein is an LDLR competitive neutralizing antigen binding protein.

    The antigen binding protein of any of the preceding claims, wherein the antigen binding protein has at least one of the following characteristics:(a) binds to a PCSK9 variant that has a D374Y point mutation;(b) is selected from the group consisting of a human antibody, a humanized antibody, chimeric antibody, a monoclonal antibody, a polyclonal antibody, a multispecific antibody, a recombinant antibody, an antigen-binding antibody fragment, a single chain antibody, a diabody, a Fab fragment, an F(ab)2 fragment, an IgG1 antibody, an IgG2 antibody, an IgG3 antibody, and an IgG4 antibody;(c) binds to PCSK9 with a Kd that is smaller than 1 nM, is smaller than 100 pM, is smaller than 10 pM, or is less than 5 pM;or(d) can block the binding of D374Y PCSK9 to LDLR with an IC50 of less than 200 pM.

    A kit for the treatment of cholesterol related disorders comprising the composition of Claims 18 or 19.

    The antigen binding protein of any one of claims 1 to 13 for use for treating or preventing a condition associated with elevated serum cholesterol levels in a subject.
