Amino acid substituted molecules

Abstract

The invention provides compositions and methods of identifying, modifying and producing modified target molecules, including therapeutic molecules by modification with non-natural amino acids. Certain aspects of the invention include methods of adding a chemical moiety to a target molecule, and the compositions resulting therefrom. Certain aspects of the invention also relate to kits for identifying, modifying and producing modified target molecules described herein.

    A method for producing a modified target polypeptide, comprising (a) providing a host cell, the host cell comprising a vector comprising a polynucleotide encoding the modified target polypeptide, and (b) growing the host cell under conditions such that the host cell expresses the modified target polypeptide, wherein the modified target polypeptide has been modified to incorporate one or more non-natural amino acids by:(i) substituting all of a particular selected amino acid residue in the polypeptide with a different naturally occurring amino acid residue;and (ii) retaining one or more of said selected amino acid residues at a non-natural position in the polypeptide and substituting said residue(s) with a non-natural amino acid residue;and (iii) optionally substituting other amino acids that interact with the substituted amino acids;and (iv) when position 1 of the polypeptide is to be substituted with a non-natural amino acid, optionally substituting position 2 in order to increase retention or removal of the amino acid substitution at position 1 and wherein the modified polypeptide retains its native function.

    The method of any of claims 1 to 3 wherein the particular selected amino acid residue to be substituted is methionine.

    The method of any of claims 1 to 4 further comprising attaching a chemical moiety to one or more of the non-natural amino acid residues in the modified target polypeptide.

    The method of any of claims 1 to 5 wherein (a) the target polypeptide is selected from the group consisting of:an antibody, antibody fragment, antibody derivative, Fab, Fab', F(ab)2, Fd, Fv, ScFv, diabody, tribody, tetrabody, dimer, trimer or minibody, a cytokine, a transcriptional modulator that modulates cell growth, differentiation or regulation, expression activator, inflammatory molecule, growth factor, growth factor receptor, and oncogene product;or (b) the target polypeptide is selected from the group consisting of: Factor VII, Factor VIII, Factor IX, Follitropin, thrombopoeitin, erythropoietin, human growth hormone, G-CSF, GM-CSF, interferon-alpha, interferon-beta, interferon-gamma, interferon-omega, interferon-tau, and GLP-1.

    The method of any of claims 1 to 6 wherein the host cell is an auxotroph for the selected amino acid.

    A modified target polypeptide wherein the target polypeptide has been modified to incorporate one or more non-natural amino acids by:(i) substituting all of a particular selected amino acid residue in the polypeptide with a different naturally occurring amino acid residue;and (ii) retaining one or more of said selected amino acid residues at a non-natural position in the polypeptide and substituting said residue(s) with a non-natural amino acid residue;and (iii) optionally substituting other amino acids that interact with the substituted amino acids;and (iv) when position 1 of the polypeptide is to be substituted with a non-natural amino acid, optionally substituting position 2 in order to increase retention or removal of the amino acid substitution at position 1;and wherein the modified polypeptide retains its native function.

    A modified target polypeptide according to any of claims 8 to 11 comprising a chemical moiety attached to one or more of the non-natural amino acid residues in the modified target polypeptide.

    A modified target polypeptide according to any of claims 8 to 12 wherein (a) the target polypeptide is selected from the group consisting of:an antibody, antibody fragment, antibody derivative, Fab, Fab', F(ab)2, Fd, Fv, ScFv, diabody, tribody, tetrabody, dimer, trimer or minibody, a cytokine, a transcriptional modulator that modulates cell growth, differentiation or regulation, expression activator, inflammatory molecule, growth factor, growth factor receptor, and oncogene product;or (b) the target polypeptide is selected from the group consisting of: Factor VII, Factor VIII, Factor IX, Follitropin, thrombopoeitin, erythropoietin, human growth hormone, G-CSF, GM-CSF, interferon-alpha, interferon-beta, interferon-gamma, interferon-Omega, interferon-tau, and GLP-1.

    A composition comprising the polypeptide of any one of claims 8 to 13, and a pharmaceutically acceptable diluent or excipient.
