Abstract

The invention provides reagents and methods for inhibiting or enhancing interactions between proteins in hematopoietic cells and other cells involved in the mediation of an immune response. Reagents and methods provided are useful for treatment of a variety of diseases and conditions mediated by immune system cells.

    A fusion peptide or peptide analogue thereof for modulating the binding of a PDZ protein and a PL protein comprising:(i) a transmembrane transporter amino acid sequence, (ii) an optional, flexible polylinker peptide sequence, and (iii) a carboxyl terminal peptide sequence comprising a PDZ domain binding motif at the C-terminal, the PDZ domain binding motif sequence comprising: E/K-X-X-V, wherein X is any amino acid, or X*-S/T-D/E-V/I/L, wherein X* is any non-aromatic amino acid.

    The fusion peptide or peptide analogue of either of claims 2 or 3, wherein the C-terminal sequence comprises the sequence E-S-D-V.

    The fusion peptide or peptide analogue of any of claims 1 to 7, wherein the carboxyl terminal peptide sequence comprises from 4 to 15 amino acids.

    The fusion peptide or peptide analogue of any of claims 1 to 7, wherein the transmembrane transporter amino acid sequence is selected from (a) tat derived from HIV, (b) antennapedia from Drosophila, (c) VP22 from herpes simplex and (d) complementarity-determining regions (CDR) 2 and 3 of anti-DNA antibodies.

    The fusion peptide or peptide analogue of any of claims 1 to 7, wherein the transmembrane transporter amino acid sequence is a tat peptide comprising the sequence GYGRKKRRQRRRG.
