Abstract

The present invention relates to methods for controlling pest infestation using double stranded RNA molecules. The invention provides methods for making transgenic plants that express the double stranded RNA molecules, as well as pesticidal agents and commodity products produced by the inventive plants.

    A method for controlling insect pest infestation, comprising providing an insect pest with plant material comprising a polynucleotide sequence that inhibits the activity of a carrier protein that is involved in the intracellular protein transport, wherein said polynucleotide sequence is selected from the group consisting of:(i) a polynucleotide sequence comprising the nucleic acid sequence set forth in SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof,(ii) a functional fragment comprising at least 15 contiguous bases of any one of SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof,(iii) a polynucleotide sequence having at least 70% sequence identity as determined using the BLASTN alignment tool to the nucleic acid sequence set forth in any one of SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof, and(iv) a double-stranded ribonucleotide sequence produced from the expression of a polynucleotide sequence of any one of (i) to (iii), wherein said polynucleotide sequence inhibits the activity of a carrier protein that is involved in the intracellular protein transport in said insect pest.

    A method for controlling insect pest infestation by providing dsRNA molecules in the diet of said insect pest or by spraying a plant surface with dsRNA molecules, wherein the sequence of the dsRNA comprises a polynucleotide selected from the group consisting of:(i) a polynucleotide sequence comprising the nucleic acid sequence set forth in SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof,(ii) a functional fragment comprising at least 15 contiguous bases of any one of SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof,(iii) a polynucleotide sequence having at least 70% sequence identity as determined using the BLASTN alignment tool to the nucleic acid sequence set forth in any one of SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof, and(iv) a double-stranded ribonucleotide sequence produced from the expression of a polynucleotide sequence of any one of (i) to (iii), wherein said polynucleotide sequence inhibits the activity of a carrier protein that is involved in the intracellular protein transport in said insect pest.

    An isolated polynucleotide sequence that inhibits the activity of a carrier protein that is involved in the intracellular protein transport selected from the group consisting of:(i) a polynucleotide sequence comprising the nucleic acid sequence set forth in SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof,(ii) a functional fragment comprising at least 15 contiguous bases of any one of SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof, and(iii) a polynucleotide sequence having at least 70% sequence identity as determined using the BLASTN alignment tool to the nucleic acid sequence set forth in any one of SEQ ID NOs 1073, 1577 to 1581, 3, 56, 168 to 172, 795, 829 to 832, 868 to 872, 890, 958 to 963, 1026, 1046 to 1050, 1684, 1771 to 1774, 2045 to 2049, 2366, 2418 to 2420, and 2466 to 2470, or the complement thereof.
