  1. 1
A method of producing an alkane or alkene from a genetically modified host cell, the method comprising:(a) transforming a host cell to express by introducing (i) an aldehyde biosynthetic polynucleotide that encodes an aldehyde biosynthetic polypeptide, and (ii) an alkane or alkene biosynthetic polynucleotide that encodes an alkane or alkene biosynthetic polypeptide;and (b) culturing said genetically modified host cell in a culture medium containing a carbon source under conditions effective to produce an alkane or alkene in the culture medium, wherein said aldehyde biosynthetic polypeptide has acyl-ACP reductase activity and is from a cyanobacterium, and wherein said alkane or alkene biosynthetic polypeptide has decarbonylase activity and is from a cyanobacterium.
  2. 5
The method of any one of claims 1-4, wherein the alkane or alkene is secreted by the host cell.
  3. 6
The method of any one of claims 1-5, wherein the alkane or alkene is a C 13 -C 21 alkane or alkene.
  4. 7
The method of any one of claims 1-6, wherein the alkane or alkene is selected from the group consisting of tridecane, methyltridecane, nonadecane, methylnonadecane, heptadecane, methylheptadecane, pentadecane, methylpentadecane, pentadecene, heptadecene, methylpentadecene, and methylheptadecene.
  5. 8
The method of any one of claims 1-7, further comprising culturing the host cell in the presence of at least one biological substrate for the polypeptide or for a polypeptide encoded by the nucleotide sequence.
  6. 9
A cell culture comprising a genetically modified host cell, wherein (a) an aldehyde biosynthetic polynucleotide that encodes an aldehyde biosynthetic polypeptide;and (b) an alkane or alkene biosynthetic polynucleotide that encodes an alkane or alkene biosynthetic polypeptide are incorporated and overexpressed in said genetically modified host cell and an alkane or alkene is produced in the cell culture when said genetically modified host cell is cultured in a culture medium containing a carbon source under conditions effective to express said aldehyde biosynthetic polypeptide and said alkane or alkene biosynthetic polypeptide, wherein said aldehyde biosynthetic polypeptide has acyl-ACP reductase activity and is from a cyanobacterium, and wherein said alkane or alkene biosynthetic polypeptide has decarbonylase activity and is from a cyanobacterium.
  7. 12
The cell culture of any one of claims 9 to 11, wherein the alkane or alkene is a C 13 -C 21 alkane or alkene.
  8. 13
The cell culture of any one of claims 9 to 12, wherein the alkane or alkene is selected from the group consisting of tridecane, methyltridecane, nonadecane, methylnonadecane, heptadecane, methylheptadecane, pentadecane, methylpentadecane, pentadecene, heptadecene, methylpentadecene, and methylheptadecene.
  9. 14
The cell culture of any one of claims 9 to 13, comprising an alkane or alkene having a δ 13 C of about -15.4 or greater.
  10. 15
The cell culture of any one of claims 9 to 14, comprising an alkane or alkene having a f M 14 C of at least about 1.003.
  11. 16
A genetically modified host cell, wherein (a) an aldehyde biosynthetic polynucleotide that encodes an aldehyde biosynthetic polypeptide;and (b) an alkane or alkene biosynthetic polynucleotide that encodes an alkane or alkene biosynthetic polypeptide are incorporated into said genetically modified host cell and an alkane or alkene is produced in the cell when said genetically modified host cell is cultured in a culture medium containing a carbon source under conditions effective to express said aldehyde biosynthetic polypeptide and said alkane or alkene biosynthetic polypeptide, wherein said aldehyde biosynthetic polypeptide has acyl-ACP reductase activity and is from a cyanobacterium, and wherein said alkane or alkene biosynthetic polypeptide has decarbonylase activity and is from a cyanobacterium.
