    A method of assaying cyclization or decyclization of a nucleotide in a sample, comprising:contacting a nucleotide with a binding partner that binds specifically to cyclized nucleotide or to noncyclized nucleotide, but not both;and detecting a response indicative of binding, or of the extent of binding, between the cyclized or noncyclized nucleotide and the binding partner, characterised in that the binding partner includes a metal ion that is required for binding between the binding partner and the cyclized or noncyclized nucleotide.

    A method of any preceding claim which is a method of assaying the activity of an enzyme that cyclizes or decyclizes the nucleotide, further comprising:contacting the nucleotide with the enzyme;and correlating the response with the activity of the enzyme.

    The method of any preceding claim, where the assay is a luminescence intensity assay, a luminescence polarization assay, or a luminescence resonance energy transfer assay.
