Compounds and methods for therapy and diagnosis of lung cancer

Abstract

Compounds and methods for the treatment and diagnosis of lung cancer are provided. The inventive compounds include polypeptides containing at least a portion of a lung tumour protein. Vaccines and pharmaceutical compositions for immunotherapy of lung cancer comprising such peptides, or DNA molecules encoding such polypeptides, are also provided, together with DNA molecules for preparing the inventive polypeptides.

    An isolated polynucleotide molecule comprising a polynucleotide sequence selected from the group consisting of:a. the nucleotide sequence provided in SEQ ID NO: 171, or the complement thereof;andb. nucleotide sequences that hybridize to the sequence of (a) under moderately stringent conditions.

    A method for detecting lung cancer in a patient, comprising:(a) contacting a biological sample obtained from the patient with a binding agent which is capable of binding to a polypeptide, the polypeptide comprising at least an immunogenic portion of a lung protein or a variant thereof, wherein said protein comprises an amino acid sequence encoded by a polynucleotide molecule comprising the polynucleotide provided in SEQ ID NO: 171;and(b) detecting in the sample a protein or polypeptide that binds to the binding agent, thereby detecting lung cancer in the patient.

    A method for monitoring the progression of lung cancer in a patient, comprising:(a) contacting a biological sample obtained from the patient with a binding agent that is capable of binding to a polypeptide, said polypeptide comprising at least an immunogenic portion of a lung protein or a variant thereof, wherein said protein comprises an amino acid sequence encoded by the polynucleotide provided in SEQ ID NO: 171;and;(b) determining in the sample an amount of a protein or polypeptide that binds to the binding agent;(c) repeating steps (a) and (b);and(d) comparing the amount of polypeptide detected in steps (b) and (c) to monitor the progression of lung cancer in the patient.

    A monoclonal antibody that binds to a polypeptide comprising an amino acid sequence set forth in SEQ ID NO:172 or the amino acid sequence encoded by the polynucleotide provided in SEQ ID NOs: 171.

    A method for detecting lung cancer in a patient comprising:(a) contacting a biological sample obtained from the patient with at least two oligonucleotide primers in a polymerase chain reaction, wherein at least one of the oligonucleotides is specific for the polynucleotide provided in SEQ ID NO: 171, or the complement thereof;and(b) detecting in the sample a polynucleotide sequence that amplifies in the presence of the oligonucleotide primers, thereby detecting lung cancer.

    The kit of claims 25 or 26 wherein the detection reagent comprises a reporter group, preferably selected from the group consisting of radioisotopes, fluorescent groups, luminescent groups, enzymes, biotin and dye particles, conjugated to a binding agent, preferably selected from the group consisting of anti-immunoglobulins, Protein G, Protein A and lectins.

    A method for detecting lung cancer in a patient, comprising:(b) contacting a biological sample obtained from the patient with an oligonucleotide probe specific for the polynucleotide molecule provided in SEQ ID NO: 171, or the complement thereof;and(c) detecting in the sample a polynucleotide sequence that hybridizes to the oligonucleotide probe, thereby detecting lung cancer in the patient.

    A diagnostic kit comprising an oligonucleotide probe specific for the polynucleotide molecule provided in SEQ ID NO:171, or the complement thereof.
