Abstract

Apparatus for handling cellular entities comprises a first substrate having an array of first wells open to a first major surface of the first substrate, said first wells being adapted to hold a cellular entity, the apparatus further comprises fluidic channels open to each well. The wells are tapered to locate the cellular entity at a given location in each well, and the fluidic channels are formed on the major surface of a further substrate adapted and arranged to face a major surface of said first substrate, the further substrate being releasably secured to said first substrate.

    A device for culturing and/or maturing a cellular entity, comprising a substrate (10, 150) having one or more wells (50) open to a first major surface of the substrate, said one or more wells being adapted to hold a cellular entity, and one or more fluidic channels (52) open to the one or more wells, the device being configured to allow exchange of liquid in the well, characterized in that the well is adapted to allow dispense and removal of the cellular entity using dispensing means, and the device is adapted to prevent the cellular entity from leaving the one or more wells through the one or more fluidic channels.

    A device as claimed in any preceding claim including retaining means (164) to retain the cellular entity against movement with the flow of liquid through the well and fluid channel.

    A device as claimed in any preceding claim in which the one or more wells is tapered to become smaller with distance from the major surface.

    A device as claimed in any preceding claim in which the one or more well is partially defined in a layer of porous material.

    A device as claimed in any preceding claim in which the one or more fluidic channels is formed by laminating a layer of porous material between two non-porous layers.

    A device as claimed in any preceding claim in which a dimension of the one or more wells is less than or equal to 3 mm.

    A device as claimed in any preceding claim in which the one or more fluidic channel has a minimum dimension of the order of half the minimal dimension of the cellular entity.

    A device as claimed in any preceding claim in which the one or more fluidic channels is provided with a gas-permeable region (78) for allowing removal of gas from the fluidic channel when being filled with a liquid.

    A device as claimed in any preceding claim in which said one or more fluidic channels is also open to one or more second wells in said major surface of said substrate.

    A device as claimed in any preceding claim in which the one or more fluidic channels is provided adjacent the base of the one or more wells, and a further fluid channel (64) is provided adjacent and in fluid communication with the top of the well.

    A device as claimed in any preceding claim in which at least one of said one or more fluidic channels is formed between the major surface of a further substrate (158) and a major surface of said substrate (150), the further substrate being releasably secured to said substrate.

    An apparatus for culturing and/or maturing cellular entities including:a device as claimed in any preceding claim, means to provide fluid flow to and from the device, and control means to control the fluid flow.

    Use of a device as claimed in any preceding claim in a method of culturing or maturing one or more cellular entities.

    A method of culturing and/or maturing a cellular entity, the method comprising:dispensing one or more cellular entities (14) into one or more wells (50) of a device as claimed in any one of claims 1 to 18, flowing a liquid medium into the one or more wells through said one or more fluidic channels (52), and culturing or maturing the cellular entity in the liquid medium in the device.
