Medicament microdevice delivery system with a cartridge

Abstract

A device for intradermal delivery of a reconstituted powdered medicament. The device includes a chamber (29), which is in fluid communication with a microdevice (50), e.g. microabrader or one or more microneedles. A cartridge (20) containing the powdered medicament may be located within said chamber. At least one burstable membrane (30) retains a powdered medicament within the housing.

    A dermal delivery device comprising:a housing (42, 45;80) comprising a chamber (29;79), an inlet port communicating with said chamber, and an outlet port communicating with said chamber;a dry powdered medicament;a microdelivery device (50) comprising a plurality of microneedles, selected from the group consisting of solid microneedles, hollow microneedles, microabraders and combinations thereof, and being sized to penetrate into the stratum corneum, intradermal space, or to disrupt the stratum corneum, without substantially penetrating into the underlying dermis;at least one burstable membrane (30, 32) sealingly attached to one of said ports to retain said powdered medicament within said chamber (29;79);and an adapter communicating with said inlet port and adapted to communicate with a source of a fluid diluent and/or a fluid reservoir (82;196) containing a fluid diluent, the reservoir communicating with said inlet port and adapted to deliver the fluid diluent to the chamber and to the microdelivery device, characterized by comprising a cartridge (20) which is retained within said chamber (29;79) when in use, wherein said cartridge comprises a passage (23), a fluid receiving opening and a fluid discharge opening, both openings communicating with said passage, and comprising a conduit (63, 64, 65) in fluid communication with said outlet port and said microdelivery device (50).

    The dermal delivery device of one of claims 1 - 3, wherein said microdelivery device (50) comprises one or more stratum corneum disrupting protrusions selected from the group consisting of microabraders, microblades, and microneedles.

    The dermal delivery device of one of claims 1 - 4, wherein the fluid reservoir (82, 196) is a container permanently attached to said adapter.

    The dermal delivery device of one of claims 1 - 5, wherein said adapter is a Luer connector for fluidly connecting said adapter to a syringe.

    The dermal delivery device of one of claims 1 - 6, wherein said at least one burstable membrane (30, 32) is burstable by the application of pressure thereto.

    The dermal delivery device of one of claims 1 - 7, further comprising a mechanical puncturing or piercing element (83) for puncturing at least one of said at least one burstable membrane (30, 32).

    The dermal delivery device of one of claims 1 - 8, wherein said adapter comprises an inlet tube communicating with said inlet port and said chamber (29;79) having a Luer connector adapted to receive a syringe barrel.

    The dermal delivery device as defined in one of claims 1 - 9, wherein said housing (42, 45) is formed of two releasably interconnected components and one of said components includes a generally cylindrical cartridge receiving chamber (29).

    The dermal delivery device of one of claims 1 - 10, wherein said cartridge (20) comprises opposed ends and said ends have convex surfaces and said burstable membranes (30, 32) comprise polyolefin film having a thickness of between 0,3 and 1,5 mils stretched taut over each of said convex surfaces at said opposed ends of said end bonded to said convex surfaces, sealing said passage (23) having a burst pressure of less than 5 atmospheres.

    The dermal delivery device of one of claims 1 - 13, wherein said housing (42, 45) is formed of two opposed thermoformed polymeric sheets bonded together, and wherein said cartridge (20) is encapsulated between said sheets forming said chamber (29).

    The dermal delivery device as defined in one of claims 1 - 16, wherein said thin burstable membranes (30, 32) have a burst pressure of less than 5 atmospheres.

    The dermal delivery device of one of claims 1 - 17, wherein the housing (42, 45;80) is cylindrical and further comprises an opening for receiving the cartridge.

    The dermal delivery device as defined in one of claims 1 - 18, wherein:said cartridge has opposed ends, said passage extending through said opposed ends of said cartridge generally coaxially aligned with said inlet and outlet ports of said housing;and said powdered medicament is contained in said passage, and burstable membranes covering said passage, and bonded to said opposed ends of said cartridge, sealing said passage.
